| Solanum tuberosum is the fourth largest crop in the world after wheat,rice and maize.It occupies an important position in China’s agriculture.Because of its rich anthocyanins and polyphenols,colored potatoes have higher nutritional and health value,and can be used as characteristic food.MicroRNAs is a ubiquitous non-coding RNA in plants.It is widely involved in plant growth and development,metabolic regulation and response to various stresses.miR858 may be one of the microRNAs involved in the regulation of anthocyanin synthesis about plants in existing reports,but the regulation of anthocyanin in colored potatoes has not been reported.In this study,the expression of miR858 in different parts of colored potato was studied,and its anthocyanin and other agronomic traits were determined to clarify the regulatory relationship between miR858 and anthocyanin synthesis.In order to find the target gene of miR858,the function of cloned StMYB48-like was predicted by bioinformatics analysis.These studies laid a foundation for clarifying the regulatory role of miR858 in anthocyanin synthesis in colored potatoes.The main results were as follows:1)The color potatoes HJG and HS66 were both mid-late maturing varieties,HM was an early maturing variety.The tubers of all varieties had shallow buds,good tuber shape and neat tuber setting.The yield per plant and commercial tuber rate were about 0.5 kg and 85%respectively.The starch content of tubers ranged from 11.5 to 15.28,and the Vc content ranged from 21.20 to 41.73.2)miR858 was expressed in different tissues of three color potato varieties,but the expression of anthocyanin in different tissues was different from that of miR858.The expression of miR858 in purple potato varieties HJG and HS66 was positively correlated with the content of anthocyanin,while the expression of miR858 in red potato variety HM was negatively correlated with the content of anthocyanin,which might be related to the content of anthocyanin.The difference of gene expression in anthocyanin regulation might be determined by the variety specificity of potato.3)In indoor and outdoor cultivation environments,the expression of miR858 in stems and leaves of three colored potatoes showed the same trend as that of anthocyanin content,and the expression of miR858 was negatively correlated with anthocyanin content in different environments.4)The same sequence of StMYB48-like gene were successfully cloned from three colored potato leaves.The location of domain MYB-DNA-binding was 2-27,and the location of signal peptide was 40-59.StMYB48-like and StMYB48 in Potato were highly correlated with AtMYB48 in Arabidopsis.It predicted that StMYB48-like was the target gene of miR858,which was involved in plant growth,development and immunity. |
