Phellodendron amurense Rupr for or genus Rhizoma Berberis plant,remnant species,the source of cork plants,or precious wood tree species,the whole body is treasure,set protection,medicinal,economic and other values in all.As a result of extensive logging and destruction,wild berberis resources have been drastically reduced and are now national level two protected tree species.There has been a lot of research on P.amurense,but there are few reports on the development and utilization of Berberis callus.Based on the callus of P.amurense,the optimum culture conditions,effective component accumulation and antioxidant ability of induced callus were investigated.It provides some reference for protecting wild P.amurense,solving the problem of supply tension of berberine drug source and developing and utilizing its antioxidant function.1.through single factor test and response surface test for analysis and design,the optimal culture conditions of induced Ramurense callus were studied.The regression model is obtained after the multivariate regression fitting of the test data.Through the analysis of the data of its model,and in order to facilitate the experiment,the optimum conditions are set:Sucrose 18g/L,nitrogen content 1.0N,6-BA1.4mg/L,repeated 3 tests under this condition,the final induction rate is about 97%,which is basically consistent with the predicted value.2.The effects of different factors on the accumulation of alkaloids in P.amurense callus were investigated by ultrasonic method and high performance liquid chromatography(HPLC)in the extraction and determination of alkaloids in P.amurense callus produced under five different inductive culture factors.The results showed that the accumulation of berberine in the total P.amurense callus was greater than the content of jatrorrhizine,and the content of palmatine was very small,among which sucrose and nitrogen had the greatest influence on the accumulation of effective components.When selecting factor conditions,in order to accumulate more secondary metabolites in callus,the growth state,genetic stability and regeneration ability of callus under this condition should also be considered synthetically.3.Using Spectrophotometry,the Ramurense callus and bark induced by the optimum conditions were used as materials to determine their respective antioxidant activity in vitro,including:DPPH Free radical scavenging ability,iron ion reduction ability,ABTS free radical scavenging ability.Through the study,it was found that the free radical scavenging ability and reduction ability of P.amurense extract and bark extract were stronger and almost equal,and had a certain degree of antioxidant ability.
