Phellodendron amurense is the main associated species in Northeast China, the national II level protected plants and one of the three famous hardwood Northeast China and the three medicinal woody plants in China. It has a high protection and exploitation prospects, with Collection of medicinal, timber and ornamental value.But it has a deeper sleep which takes a lot of inconvenience to the production of artificial propagation. In this study, the Phellodendron amurense seed as experimental material,according to its own characteristics dormancy, germination rate and other characteristics. We have a study of its dormancy releasing method, and the physiological change mechanism during seed dormancy breaking and other aspects. We reached the following conclusions:(1) Take the method of cold stratification, temperature stratification, cold stratification and hormone treatment to break seed dormancy. Sampling test and checking germination every 10 days, which put in low temperature 4℃ and 10℃ processing. We get the seed germination rate in 4℃ stratification seed is higher than that in 10℃ product processing layer, and the germination rate is significantly higher after changing temperature. So we think the 4℃ cold stratification is the optimum temperature to break seed dormancy.(2) Hormone treatment combined with low germination can improve rate significantly. We collected from different concentrations of GA,6-BA, NAA treatment of seeds,and found that the 6-BA100mg/L, GA500mg/L and NAA10mg/L combing with 4℃ cold stratification solution after soaking binding to break Phellodendron amurense seed dormancy effect most best, can significantly improve the germination rate of seed dormancy releasing very effective.(3) Seed germination requires energy, the metabolic activity of seed needs lot of energy contents. According to the Conclusions, take the method of 4℃ cold stratification, determined the internal inclusions changes at regular intervals. The outcome is that the soluble protein content had small increase and gradually smooth and trend.Soluble sugar content increased first and then decreased.The starch content of a substantially decreasing trend. The fat content increased first and then decreased.(4) Endogenous hormones promote or restrict the seed seed germination during seed dormancy breaking of Phellodendron amurense, take the method of Solid-Phase Extraction and HPLC to determine the content changes of the four kinds of endogenous hormones as GA, IAA, Z, ABA. Wherein, GA, IAA, and Z increased in varying degrees. But ABA declined with stratification time extended. Description during seed dormancy breaking of Phellodendron amurense. GA, IAA and Zeatin can break seed dormancy effectively and promote seed germination. The ABA restran cell growth and hinder seed germination.(5) Berberine, jatrorrhizine and palm leaf tetrandrine are not only the important secondary metabolites, but also the main medicinal ingredient of Phellodendron amurense. During seed dormancy breaking of Phellodendron amurense,the three alkali content increased in varying degrees,among that we can reach clearly that, Berberine had the highest content, followed jatrorrhizine, palm leaf tetrand had the minimum content.
