With the progress of human society, the industrialization developed repidly, impact on nature was becoming more frequently. The destruction of the ozone layer reduced ozone content in the stratosphere, directly lead to enhanced UV-B radiation. This paper was investigated the effect of different dose and treatment time of supplementary UV-B radiation on Phellodendron amurense Rupr.seeding grown in the greenhouse. In order to foud out the best way to increased synthesis of alkaloids:The dose of supplementary UV-B radiation were 9.78μw·cm-2·nm-1(higy dose),and 3.26μw·cm-2·nm-1(low dose),and the radiation time were 10d(short term)、25d(tansition period) and 40d(long term). In order to foud out the best way to increased synthesis of alkaloids. The main results can be summarized as follows:1. There were noticeable effects of supplementary UV-B radiation on biomass of P. amurense seedling (P<0.05), low doses of UV-B radiation treatment was more advantageous and the biomass growth increased 46.26%.2. There were noticeable effects of supplementary UV-B radiation to MDA and ·OH content(P<0.05).UV-B radiation could significantly increased the content of MDA and ·OH. With the extension of UV-B radiation processing time, the content of MDA was reduced, but the content of ·OH was increased. High dose of UV-B radiation was good for P. amurense seedling.3. There were noticeable effects of supplementary UV-B radiation to the tannins content (P<0.05),high doses of UV-B radiation in the treatment of the prophase and metaphase was increased 49.29%,and the low doses of UV-B radiation treatment group, the tannins content increased significantly.4. There were noticeable effects of supplementary UV-B radiation on photosynthesis and fluorescence characteristics of P.amurense seedling(P<0.05),UV-B radiation decreased the photosynthsetic rate, transpiration rate and stomatal conductance. The FV/Fm、FV/F0、ETR values were decended.5. There were noticeable effects of supplementary UV-B radiation to the secondary metabolites content which in the roots(P<0.05),stems and leaves, UV-B radiation was good for the accumulation of flavonoids. As the extension of processing time, the content of Berberine and Palamatine also increased significantly, but bad for Jatrorrhizine.Comprehensive the above analysis:UV-B radiation was bad for physiology and biochemistry of P. amurense, with the treatment time and the radiation dose, the P. amurense has certain adaptability. It has a great significance for medicinal plants. At the same time,it also has a certain significance for use the secondary metabolites.
