Hypsizygus marmoreus is a large-scale high-quality raise edible fungus with crisp texture,delicious taste,rich nutrition and health care value.It has a large market at home and abroad,and the demand and cultivation area are also increasing year by year.However,the quality of the strains in Hypsizygus marmoreus industry is uneven,the yield is low,and the resistance is poor,which affects the benefits.Therefore,the genetic breeding of Hypsizygus marmoreus will contribute to the sustainable development of the industry.In recent years,research on the molecular genetics of edible fungi has made some progress.People have a certain understanding of the mating factors of A and B.However,at present,there is little research on the genetic aspect of Hypsizygus marmoreus,and no mature theoretical system has been formed.The guidance of breeding work is not yet sufficient.The mating type control is closely related to the development of the fruiting body.The mating type identification of monokaryons is an important basis in the breeding work.Therefore,the mating type research on breeding has a very important meaning.In this paper,many protoplast monokaryons and spore monokaryons were isolated by protoplast mononuclearization and single spore isolation using B5 of a strain of Hypsizygus marmoreus.After routine analysis of the mating type of tetrapolar bacteria,the four mating types of spore monokaryons were accurately identified by nuclear migration assay.The resaults could be categorized as the following 3 tips.1.567 protoplast monokaryons were obtained by protoplast mononuclearization,and the mating types of test strains T1 and T2 were determined to be A1B1 and A2B2 by standard test strain method.2.94 spore monokaryons were obtained by single spore isolation.The number of four mating types of T1,T2,T3 and T4 was routinely identified as 24,27,25 and 17,respectively.And a recombinant mating type T5(ArBr)was also identified.3.Through the nuclear migration test,four mating types were accurately identified as A1B1,A2B2,AIB2,and A2B1.It was verified that the separation law of spore monokaryons conformed to Mendel's law of genetic separation,and it was confirmed that Hypsizygus marmoreus is a quaternary heterozygous combination basidiomycete.
