| Hypsizygus marmoreus is a kind of edible fungi, which has achieved the factory mode of production at home and abroad. The color of H. marmoreus fruit body on the market can be divided into two broad categories, to be dust-color and white.White H. marmoreus compared with dust-color H. marmoreus is higher at price.But white species is limited and has a longer planting period,low yield,weaker resistance.Therefore, it is very theoretical and practical significance to carry out germplasm research of white strain, material preparation and experimental exploration, to enrich germplasm resources of the white strain.In this paper, the relationship between the two colors was initially identified by spore monokaryons hybridization of the two kinds of color of Hypsizygus marmoreus.The specific marker of the white strain, the genetic polymorphism of its protoplast monokaryons and spore monokaryons were further explored by RAPD technology.1.27H.marmoreus strains in our Preservation Center were cultivated and their color was observed.The result showed that in addition to pure white SIEF336strain,the cap color of other strains was dust-color or deep dust-color. On this premise, SIEF3133(dust-color) and SIEF3136(pure white) were selected for material to analysize fruit-body color.67SIEF3133monokaryons and82SIEF3136monokaryons were gotten by single-spore isolation. Out of16groups,12groups were compatible by hybrid pairing. A total of48heterozygotes were received, and two fruiting color statistics showed that the color of all hybrid strains was the same as the SIEF3133’s, which was very different from the pure white of SIEF3136fruit body. So the dust-color was dominant compared with pure white.2. RAPD primers were primarily selected by two protoplast monokaryons,so clear and recognizable bands can be amplified by72primers,from which38polymorphism primers were screened among30SIEF3136monokaryons of different mating types, A total of341fragments were amplified, among them253(accounting for74.2%) were polymorphic.Genetic similarity coefficient matrix among31strains ranged from0.34to0.90.Similarity coefficient of the same mating type monokaryons was above0.60,which showed a higher similarity, all strains were divided into2groups.PM-01with T1(A3B3), T3(A3B4) was clustered into the first category,and PM-02with T2(A4B4), T4(A4B3) into the second category.In addition,out of54pairs of ssr primers to amplify SIEF3136monokaryons polymorphic bands could be amplified by21pairs.That polymorphic rate of54.86%indicated SSR markers could be used as research tool for H.marmoreus monokaryons.3. Specific bands of three primers only on SIEF3136were obtained to amplify27H.marmoreus strains by screened primers. Specific band Ts2116can be stablely transmitted in all hybrid strains and monokaryons of SIEF3133, but in all the tested monokaryons of SIEF3136, this band can not be found.So this marker can be taken as specific identification tag for SIEF3136strain. That cap color of hybrid strains was dust-color suggested that the marker may be related to the dust-color gene. |
