Chieh-qua(Benincasa.hispida Cogn.var.clhieh-qua How),also known as hairy squash,hairy Chieh-qua,little wax gourd etc.,is an annual herb of the genus Bencurasa of Cucurbitaceae.It is a melon vegetable that is widely consumed in southern China.In recent years,the area planted with Chieh-qua has been expanding,and the market for melon seeds also has gradually expanded.With the emergence of new hybrid varieties,it is more and more important to quickly identify the purity of the seeds of Chieh-qua.Seed,as the most basic means of production in agricultural production,is an irreplaceable factor in agricultural production.Therefore,seed quality inspection is very important in the process of seed production and sale,and seed purity identification is an important part of seed quality inspection.This research based on the genome sequencing results of Chieh-qua,and the SSR primers suitable for Chieh-qua were designed.By using SSR molecular marker technology,to establish a technical system for rapid indoor identification of seed purity of Chieh-qua hybrid,in order to identify the purity of Chieh-qua cultivars.The main results are as follows:1.By studying the methods of DNA extraction,we preliminarily explored the rapid DNA extraction method suitable for SSR-PCR of Chieh-qua(alkali treatment),and compared the DNA extraction method with CTAB method.2.200 pairs of SSR primers were screened from 8 parental DNA samples.Eight pairs of SSR primers with stable amplification,simple banding pattern,paternal dominance and parental co-dominance were obtained,which could be used for purity identification of 6 hybrid combinations.Sixteen hybrid seeds were identified quickly by the research team,and compared the 10 of them field identification results,to verify the accuracy of the modified technique system.3.In this study,a rapid DNA extraction method(alkali treatment)to Cotyledons of Chieh-qua was combined with SSR molecular marker technique,establishing a preliminary SSR molecular marker purity identification system for hybrid seeds of Chieh-qua.
