Amygdalus triloba f.multiplex,which belongs to Rosaceae,peach plants of the genus.has characteristics of early flowering,long flowering period,large flowers,wide geographical distribution,and resistance to dry soil,adaptability.Thus it is the main spring flowering tree in northern China.In this paper,Amygdalus triloba f.multiplex was used as the material to study polysaccharide.The main contents of the study are as follows:(1)The extraction technology of polysaccharide in Amygdalus triloba f.multiplex flowers was optimized by ultrasonic assisted extraction.The dynamic changes of polysaccharide content were then studied under the optimum conditions.The response surface methodology was used to optimize and analyze variance of the main extraction parameters.The results showed that the optimal extraction conditions were as follows:ultrasonic time was 40 min;extraction temperature was 70℃;ultrasonic power was 120 W;the solvent to material ratio was 60 mL/g.It was found that the content of polysaccharide in flowers was the highest at early blooming flowers,and the yield of polysaccharide reached 2.81%.The utilization value of early blooming flowers was relatively high.(2)Study on the separation method of polysaccharide in Amygdalus triloba f.multiplex flowers.Polysaccharides from four blooming periods of Amygdalus triloba f.multiplex flowers was separated by AKTA Purifier UPC 10 biomacromolecules purification system and Superdex 75 column chromatography,and the eluent of polysaccharide was optimized.The results showed that the ultrapure water was the best to elute the polysaccharide solution,the separation of each flowering period polysaccharide obtained three main polysaccharide components,the content of each polysaccharide was basically consistent with its growth and metabolism.(3)Determination of the molecular weight of Amygdalus triloba f.multiplex polysaccharide.Using AKTA Purifier UPC 10 biomacromolecules purification system and Superdex 75 columm chromatography,the molecular weight of three components of polysaccharide of Amygdalus triloba f.multiplex were determined.The results showed that the molecular weights of these three components were 5.17×104,9.41×103 and 3.03×103.This study lays a good foundation for the study of the relationship between the structure and the mechanism of polysaccharides of Amygdalus triloba f.multiplex. |
