Coccidiosis poses a serious threat for poultry industry. It can cause a large number of poultry deaths, reduce animal performance and result in large economic losses. The development of anticoccidial drugs has greatly promoted the development of the chicken breeding industry.Hainanmycin sodium is a kind of polyether anticoccidial drug that specially used to prevent and treat chicken coccidiosis. In addition,it is a new type of anticoccidial drug approved by China.Although this drug has been on the market for more than 20 years, its effectiveness in treating intestinal coccidia infection of chickens and the safety in targeted animals have not been systematically evaluated so far. Therefore,it is imperative to investigate the effectiveness of Hainanmycin premix on chicken intestinal coccidia and the safety evaluation in targeted animals, which contribute to enrich the clinical pharmacology research content of domestic Hainanmycin, guide clinical rational drug use and reduce the occurrence of toxic and side effects.1.Pharmacodynamics of Hainanmycin Sodium Premix on chicken coccidiaIn order to verify the effect of Hainanmycin Sodium Premix on chicken coccidia, the models of infecting artificially Eimeria acervulina,Eimeria maxima and Eimeria necatrix were replicated to evaluate effectiveness of Hainanmycin Sodium Premix on chicken coccidia at the recommended dose, and compared with Monensin and Diclazuril. All drugs were given by mixed feeding and feeding freely. The experimental period of efficacy of Eimeria acervulina was 9d,and that of efficacy of Eimeria maxima and Eimeria necatrix was 10d.The anticoccidial index against Eimeria acervulina was 177.7,and Eimeria necatrix was 175.3,both reaching intermediate-effect anti-coccidial levels; The insect index resistance to Eimeria maxima is 180.5,achieving high anti-coccidial levels, and both better than Monensin and Diclazuril. These results showed that Hainanmycin Sodium Premix had a better effect on Eimeria acervulina at the recommended dose of 7.5mg/kg for 9d,and possessed a comparable effect on Eimeria maxima and Eimeria necatrix at the recommended dose of 7.5mg/kg for 10d.2.The safety evaluation of target animalsForty 15-day-old yellow-feathered broilers were randomly divided into four groups, ten in each group, half male and female. The test medicine of Hainanmycin Sodium Premix was divided into three groups:1 times(7.5 mg/kg),1.5 times(11.25 mg/kg)and 2 times(15 mg/kg).According to the recommended route of administration, the medicine was mixed and administered for 28d.At the same time,an empty control group was established without the addition of drugs.Clinical performance, body weight and feed intake of each group of chickens during the study period were monitored;Besides, the hematological and blood biochemical parameters on the 14th and 28th days after the administration were measured. At the end of the experiment, the animal necropsy and histopathological examination were performed to evaluate the safety of target animals used in chickens with the application of Hainanmycin Sodium Premix. The results showed that there was no significant effect on the growth performance of chickens in different dosage groups of Hainanmycin Sodium Premix.Regarding the physiological and biochemical parameters, although there were significant differences in some of the parameters compared with the control, there was no significant dose-dependent change, nor any evidence demonstrated that it was related to the use of medicines.Collectively,Hainanmycin Sodium Premix is safe for targeted chicken by mixed feeding according to the recommended dose.
