Chlorella Vulgaris is a genus of single-cell green algae belonging to the phylum Chlorophyta.It is abundant in amino acids,vitamins,protein.It have widely applied in aquaculture,bio-energy production and functional health food development fields.The Growth of Chlorella Vulgaris is based on nutrients such as nitrogen and phosphorus,besides it is the accumulation of basic protein,amino acids,polysaccharides and other bioactive substances.Light,temperature,ventilation and other external training environment on Chlorella Vulgaris Growth and biological components play a decisive role.In this experiment,we based on f/2 to culture Chlorella Vulgaris,by chaging respectively the phosphorus concentration,the nitrogen concentration,temperature,brightness than the time,aeration rate to explore their effect on the growth of Chlorella Vulgaris.The main results of this paper are as follows:1.Effects of Different phosphorus concentration on growth,protein content and amino acid composition of Chlorella Vulgaris2.It was based on f/2,Na H2PO4·H2O as the phosphorus source,which were added to the phosphorus concentration of 0mg/L,5 mg/L,25 mg/L,50 mg/L,75 mg/L,100 mg/L,200mg/L of Na H2PO4·H2O.The experimental results showed when the phosphorus concentration of 50mg/L,cultured for 16 days,maximum protein content was 38.35%,23.35% higher than the initial value,when cultured for 12 days,maximum threonine content was 3.21g/100 g,when cultured for 16 days,leucine maximum value was 2.32g/100 g.2.Effects of Different nitrogen sources on growth,protein content and amino acid composition of Chlorella Vulgaris3.It was based on f/2,Na NO3?NH4HCO3?NH4NO3?NH4CI?(NH4)2S04?NH4H2PO4as the p nitrogen sources,The results showed that with Na NO3 as nitrogen source,cultured for 16 days,the growth reached a maximum OD680,which was 0.752,higher than the initial value of 0.622,urea as a nitrogen source,cultured for 20 days,the protein content of maximum was 42.10%,compared with the initial value higher was 27.10%,while urea as a nitrogen source,cultured for 20 days,threonine,serine,glycine reached the maximum content significantly higher than other nitrogen sources of culture,respectively 3.02g/100 g,2.786g/100 g,2.302g/100 g,with NH4 Cl as nitrogen source when cultured for 16 days,cysteine,isoleucine content of the highest values,significantly higher than other nitrogen content of culture,reaching 0.921 g/100 g,1.486g/100 g.3.Effects of Different nitrogen concentration on growth,protein content and amino acid composition of Chlorella VulgarisIt was based on f/2,Na NO3 as the nitrogen source,which were added to the nitrogen concentration of 0.000 g/L ?0.0375g/L?0.075g/L?0.3g/L?0.6 g/L?1.2 g/L?2.4 g/L of Na NO3.The experimental results showed that with Na NO3 as nitrogen source,when the nitrogen concentration of 0.6g/L,cultured for 16 days,the growth reached a maximum OD680,which was 0.752,0.622 higher than the initial value,maximum protein content 40.5%,higher than the initial value of 25.5 %,when the nitrogen concentration was 0.075g/L,cultured for 16 days,alanine,glutamic acid,serine content of the highest values,significantly higher than other nitrogen content of training,respectively,2.265g/100 g,3.952g/100 g,2.012g/100 g,when the nitrogen concentration of 0.6g/L,cultured for 20 days,lysine,methionine content of the highest values,significantly higher than other nitrogen content of culture,reaching 4.012 g/100 g,4.213g/100 g,when the nitrogen concentration of 0.3g/L,cultured for 20 days,reached the highest value tyrosine content 2.482g/100 g,significantly higher than other nitrogen content of culture4.Effects of Different temperatures on growth,protein content and amino acid composition of Chlorella Vulgaris5.It was based on f/2,Chlorella Vulgaris were placed respectively in 15?,20?,25?,constant light incubator 30?,35? in static culture,grown OD680..The results showed when temperature was 25?,cultured for 16 days,OD680 was 0.752,which reached the maximum,0.622 higher than the initial value,when the temperature is 30?,cultured for 16 days,the protein content of maximum is 44.35%,29.35% higher than the initial value.when the temperature was 30?,cultured for 8 days,tyrosine content reached the highest value,which was 2.724g/100 g,significantly higher than in other temperatures;When cultured for 20 days,aspartate peaked was 4.482g/100 g,significantly higher than other temperaturest.5 Effect of Different Light-dark on Growth,protein content and amino acid composition of Chlorella VulgarisIt was based on f/2,setting brightness ratio(Light: Dark = L: D)was 0: 24,12: 12,14:10,24: 0.The results found that when dimming ratio of 0:24 and 24: 0,Chlorella growth were inhibited,the growth OD680 continued to decrease over time,when the brightness ratio of12:12 and 14:10,growth OD680 was increasing over time,after cultured to 16 days,the growth reached maximum,which OD680 was 0.798 and 0.752,the highest protein content values were42.2% and 40.5%,between the two dimming ratio,the difference was not significant,but significantly higher dimming ratio of 0:24 and 24: 0.6 Effect of Different aeration rate on Growth,protein content and amino acid composition of Chlorella VulgarisIt was based on f/2,the ventilation rate were 0L/min,1L/min,2L/min,3L/min.The results found when the aeration rate was 1L/min,cultured for 16 days,the growth is 0.920,which reached a maximum OD680,0.790 higher than the initial value,the maximum value of the protein content was 45.3%,30.3% higher than the initial value,tyrosine,glutamic acid,maximum content of serine,respectively in an amount of 3.214g/100 g,5.876g/100 g,4.523g/100 g.
