| In this study,the changes of reproductive hormones in the breeding cycle of Yili goose were measured,combined with the observation of the development of ovary and follicular,to explore the changes of reproductive hormones in reproductive cycle of Yili goose;through exogenous hormonal regulation,determination of reproductive hormones,ovaries and follicles in the nest period Yili goose;detection of polymorphism of laying performance related genes of Yili goose,by using PCR-SSCP technology.Through the above research to provide a theoretical basis for improve the reproductive performance,expand its populationand and study the reproductive control techniques and molecular marker assisted selection of Yili goose.Experiment 1:2 age Yili geese were chosen which were in good health,similar weight.32 samples were randomly selected to take blood samples for hormone detection during the laying period、broody period and ceased period.And 5 samples were randomly selected to gather ovarian tissue for the observation of the development of ovary and follicular.The results showed that:during the laying period,the content of GnRH of the Yili goose was significantly higher than the broody and ceased period by16.70%(P<0.01)and 58.28%(P<0.01);the PRL was significantly lower than the broody period by13.00%(P<0.05),significantly higher than the ceased period by 28.94%(P<0.01);the content of FSH was higher than the broody period by 6.98%(P>0.05),Significantly higher than the ceased period21.12%(P<0.0l);the content of LH was significantly lower than the broody period by 8.38%(P<0.0l),significantly higher than the ceased period of 16.84%(P<0.05);the content of E2 were significantly higher than the broody and ceased period of 29.80%(P<0.0l)and 112.40%(P<0.0l);the content of P4 were significantly higher than the broody and ceased period by 28.89%(P<0.0l)and 30.34%(P<0.0l).The volume and weight of ovaries of Yili goose were significantly higher than the content of broody and ceased period(P<0.01).The ovaries multi-grade follicle growth and development were active and structured in laying period;on the broody period,the internal ovaries were full of different sizes of follicles which were atrophy and atresia;on the ceased period,the level of ovaries were more atrophy than the ceased period,the cytoplasmic cavity of the follicle becomes larger and tend to inyagination.Experiment 2:2 age Yili goose 30 were chosen which were in good health and similar weight were randomly divided into 5 groups.The control group was blank control and intramuscular injection of saline group,the test group is consisted of intramuscular injection of testosterone propionate group,intramuscular injection of LHRH-A3 group and oral methanesulfonic acid bromocriptine group.At 1 d,3 d,5 d,7 d,10o’clock in each trial,each fasting wing vein blood collection 4 mL,related reproductive hormone was measured.Test Day 14 d,4 samples were randomly selected to gather ovarian tissue for the observation of the development of ovary and follicular in each group.Results show that:The GnRH and PRL of Yili goose plasma were on the decline,and the increasing trend of FSH,LH,E2 in fluctuation change,P4 concentration changed little,after intramuscular injection testosterone propionate processing.The ovaries weight,width and thickness of experimental group was increased than that of the control group 81.40%(P<0.05)、57.93%(P<0.05)and 41.65%(P<0.05).The LH and GnRH of test group presented the first rise after falled,after intramuscular injection LHRH-A3 processing,the ovary volume and weight of Yili goose were higher than those of control group(P<0.01),there was no significant difference;The width of ovaries was higher than that of the control group 66.19%(P<0.05),and The thickness of ovaries was higher than that of the control group 62.99%(P<0.01).The PRL,P4,E2 of the Yili goose was significantly decreased,and FSH was significantly increased,after oral bromine hidden pavilion mesylate tablets.The follicular diameter,oocyte diameter and granular cell layer thickness of primordial follicle of experimental group was respectively higher than that of the control group 34.43%(P<0.05)、35.00%(P<0.05)and 50.00%(P<0.05);The follicular diameter,oocyte diameter and granular cell layer thickness of primary follicle of experimental group was respectively higher than that of the control group 24.68%(P<0.05)and 30.09%(P<0.05).Experiment 3:68 2 age Yili goose were chosen to take blood samples,gene polymorphism of GnRH 5’regulatory area,PRL-exon5,ESR-exon4 and FSHβ-exon2 had been detected by PCR-SSCP.The result shows:two genotypes of GnRH gene were detected,as AA,AB,genotype frequency of GnRH gene in this group were 0.70 and 0.30,respectively;three genotypes of FSHβgene were detected,as AA,AG and GG,genotype frequency of GnRH gene in this group were 0.53,0.10 and 0.36,respectively.ESR gene exon 4and PRL gene exon 5 were detected for polymorphism.The polymorphism information content(PIC)of GnRH and FSHβgene were 0.222 and 0.367. |
