Production Of Compound Chinese Medicine Ointments For Inactivate The Superficial Fungal On Animal And Medicine Efficacy

Dermatomycosis is a zoonosis,with wide range,high morbidity,fast transmission,serious damages,which is caused by mycoses of superficial fungi.Chemically-synthesized medicine was widely used to cure dermatomycosis clinically,however,which can induce side effects like narrow antimicrobial spectrum,drug-resistant strains,toxicity and reduce immunity.In this study,the HPLC determination conditions for Chinese medicine extracts instructions composition and production of compound Chinese medicine ointments were explored,and the artificial Trichophyton mentagrophytes(T.mentagrophytes)infection animal model has been established to proceed efficacy test,skin irritation and acute toxicity test.The results are shown as following:1.Using HPLC to content determination for Cortex Pseudolaricis,cortex dictamni,sophora extract of compound Chinese medicine instructions composition,and the determination of each ingredient required for chromatographic conditions were determined,respectively:(1)Fraxinellone,obakunone,Pseudolaric acid B:Agilent C18(250 nm×4.6 nm,5 ?m),Mobile phase:CH3CN-H2O(2‰ H3PO4),Time and proportion:0-30min,CH3CN 30%-80%;30-51min,CH3CN 80%-30%;Speed:1.0mL/min;Temperature:25?;UV detection wavelength:Fraxinellone,obakunone 236 nm,Pseudolaric acid B 260 nm.(2)Oxymatrine:Agilent ZORBAX NH2(5?m,4.6×250mm);Mobile phase:CH3CN-CH3CH2OH4(80:20);Speed:1.0mL/min;Temperature:27.7?;UV detection wavelength:220nm.(3)Fraxinellone 11.54 ?g/mL,stability and repeatability,RSD 1.22%,1.25%;average recovery rate 100.26%,RSD 0.23%.2.Using orthogonal experiment design to screen the best cream matrix formula,prepare compound Chinese medicine ointments,and conduct stability test,and determine the instructions in the paste composition with HPLC.The best ingredient is dertermined as:lauryl sodium sulfate 1.55g,glycerin monostearate 3.45g,propylene glycol 8mL,ethylparaben 0.3g,stearic acid 6.8g,Vaseline 6.8g,liquid paraffin 3.4g,azone 2.5mL,Extract ingredients stability,the composition of indicators can be detected.3.Establishing the artificial Trichophyton mentagrophytes(T.mentagrophytes)infection animal model to proceed efficacy test,and evaluating the efficacy of Chinese medicine ointment compound through clinical disease score,tissue culture,and histopathological observation.Results showed that disease symptoms of treated groups disappeared after using drug 22 days.According to the Veterinary Drug Research Technical Guiding Principles as effecttly,the therapeutic effects in high Chinese medicine ointment compound groups,middle Chinese medicine ointment compound groups and terbinafine groups are basically the same,and the therapeutic effects in the low Chinese medicine ointment compound groups and liquid compound are similar.4.According to the study of Chinese medicine,natural medicine topo-irritation and hemolytic technical guiding principles,the reaction of skin irritation and acute toxicity on the rabbit health skin and damaged skin after using the compound Chinese medicine ointment were observed.Results showed that the compound Chinese medicine ointment was nonirritant for health skin,mild irritated for damaged skin,and non-poisonous.
