Objective: Female infertility can lead to heavy losses of China's animal husbandry production.One of important reasons for infertility is that the animal had endometritis.The pathogenesis of endometritis is complex and needs further study.As a predictive factor for inflammatory diseases,asymmetric dimethylarginine(ADMA)can promote endometrial dysfunction.However,it has not been reported that ADMA is associated with endometritis.Therefore,rat model of endometritis was used to detect the changes of ADMA content in the rat uterus homogenate,and to clarify the mechanism by detecting the expression of ADMA-related enzymes in the present study.Methods: The 90 Wistar rats in preliminary experiment were randomly divided into five groups.0 mg/mL,1 mg/mL,3 mg/mL,5 mg/mL,and 7 mg/mL 60 ?L LPS were injected to establish a rat model of endometritis.By the clinical examination,pathological sections,and the relative fluorescence quantitative PCR detecting the gene expression level of tumor necrosis factor alpha(TNF-?)and interleukin 6(IL-6)in the uterus homogenate of rat were used to analyze and appraise it.5 mg/mL was determined as the best dose.36 Wistar rats in the formal experiment were randomly divided into two groups: one 5mg/mL 60 ?L LPS treated group and another 60 ?L normal saline injected group.The content of ADMA in uterus was detected by HPLC.The relative fluorescence quantitative PCR was used to detect the expression level of ADMA synthase(protein arginine methyltransferases,PRMT)and ADMA decomposition enzyme(dimethylarginine dimethyl-aminohydrolase,DDAH)in uterine homogenate.Results:The results showed the rats have a significant loss of appetite.These rats liked to lie down and liked to be quiet after the attack of 24h;A large number of eosinophils and the phenomenon of falling out of the endometrium were observed in the pathological sections.The levels of TNF-? and IL-6 were significantly higher in the experimental group(5 mg/mL)than in the blank control group(P < 0.01).The results showed the content of ADMA in uterine homogenate of endometritis rats was greatly increased compared with the blank control group(P < 0.01).The results showed the expression of PRMT1 in the uterus of endometritis rats was significantly higher and the expression level of DDAH1 was significantly lower than in the control group(P < 0.01).Conclusions: The occurrence and development of endometritis in rats is related to the increase of ADMA content.The increase in ADMA content is related to the up-regulation of PRMT1 expression and down-regulation of of DDAH1.
