Objective: Postoperative pain is one of the key factors affecting postoperative recovery,if not timely scientific intervention,a serious threat to the health of the body,and even fatal.But the postoperative pain involving a large number of substances,the mechanism is unknown,the current can't be an objective assessment,but can't scientific intervention.Thereforethis study is based on the postoperative pain model of rats,the use of metabolomics method of comparative study of rat hypothalamus in the differential metabolites,in order to find postoperative pain biomarkers and elucidate the mechanism of postoperative pain and objective assessment and scientific Intervene to lay the foundation.Methods: The fracture model of rat left hind limb was established by referring to Brennan postoperative pain model.Von Frey electronic pain meter records the rat mechanical compression threshold.The metabolites of hypothalamus were detected by GC-MS.The differential metabolites were mapped to K EGG database.The metabolic pathway of KEGG ID was used to analyze the metabolites of differential metabolites.Res?Lts: The res?Lts showed that the amount of activity after fracture was significantly reducedand the left hind limbs were not burdened.The food intake was significantly reducedand the lobes and bite fractures were completely lifted and the hair was dim and d?Ll.Von Frey electronic pain measuring instrument records: 0.5 h after surgeryleft hind limb mechanical compression threshold was 3.21 ± 0.14 g,2 h after surgery was 4.46 ± 0.11 g,6h after surgery was 4.93 ± 0.13 g,12h after surgery 6.69 ± 0.14 g,10.70 ± 0.14 g after 24 h,compared with the preoperative difference was significant,statistically significant(p<0.05).The sensitivity of the method was 79.33%,the specificity was 100%,the Gordon index was 0.9333,and the coincidence rate was 96.66%.Based on the rat model of fracture and pain,the metabolomics analysis of GC-MS was used to select 1-methylheptene,3-aminoisopropionic acid,acetanilide,cystine,benzene Formic acid and other 16 kinds of different metabolites.Conclusion: The postoperative pain model of rat fractures was established successf?Lly.Based on the model,16 different postoperative pain metabolites were successfully screened.It is important to find a mechanism for postoperative pain biomarkers and to elucidate the mechanism of postoperative painas well as objective assessment and scientific intervention.It is important to promote the minimization of animal pain.
