Uncaria rhynchophylla to test materials for the study of the influence of different environmental conditions on the growth and development of U.rhynchophylla.First,discussing the significant factors of U.rhynchophylla seed germination and pretreatment method of U.rhynchophylla seeds;Second,One-year-old U.rhynchophylla seedling were used to the test materials,and exploring the influence of different light intensity on seedling growth of U.rhynchophylla;Finally,three years U.rhynchophylla seedling were used to the test materials,the effects of altitude on plant morphology,physiology,biochemistry and growth and development were preliminarily studied.Meanwhile,it was analyzed that the correlation between altitude and plant morphology,physiological and biochemical indexes.The main results were as follows:1?The seed germination rate had a significant impact on different storage temperature,light,matrix water content and pH.The optimal storage temperature of U.rhynchophylla seed was 4?,under this condition,the germination rate,germination energy and germination index were the highest.The germination rate and germination index were higher in 10001500 lx light intensity;50%of the matrix water content is more suitable for the germination of the seeds;When pH was 6.5,weak acid environment was more suitable for U.rhynchophylla seed germination.In the pretreatment methods such as GA3,KNO3,hot water soaking and H2SO4,etc,the highest germination rate of U.rhynchophylla seed was treated with 100 mg·L-1GA3.2?The effects of different light intensity on the seedlings growth of U.rhynchophylla were studied by shading facilities.The results showed that the light shading were conducive to the organs growth of U.rhynchophylla seedlings,and various morphological indexes?e.g.plant height,stem diameter and branch number?have been more significantly improved relative to full illumination,the moderate shading and the severe shading.The net photosynthetic rate,soluble protein content and soluble sugar content under four light conditions showed full sun light>light shading>medium shading>severe shading.The leaf water content and the content of total chlorophyll increased with the increase of shade density.The stomatal conductance of leaf showed light shading>full sun light>medium shading>severe shading.MDA content of leaf showed light shading<medium shading<full sun light<severe shading.Analyzing synthetically morphological indexes,physiological and biochemical indexes of U.rhynchophylla seedlings under four light conditions,we thought that shading of 55%75%were more conducive to the seedlings growth of U.rhynchophylla.3??1?Environmental conditions in different planting test areas:About altitude,Danzhai?820830 m?>Changshun?10201070 m?>Guiyang?11301150 m?>Pingba?12301240 m?.The soil of four planting test areas is acidic soil,and the pH value is between 5.5 and6.5;The contents of soil organic matter,total nitrogen,alkali hydrolyzable nitrogen,total phosphorus,available phosphorus,total potassium and available potassium in four planting test areas were not significantly different.?2?Regular detection of morphological indexes,physiological and biochemical indexes of U.rhynchophylla plants at various altitudes region showed that:In four altitude cultivated fileds,in general,the morphological indexes?e.g.plant height,stem diameter,branch length,branch number,number of branches,number of nodes,numbers of total leaves and hooks?,physiological and biochemical indexes?e.g.leaf photosynthetic parameters,photosynthetic pigment content,soluble sugar and soluble protein?in the planting area of 820830 m?Danzhai?was obviously higher than other planting areas.In addition,altitude was negatively correlated with morphological indexes and physiological and biochemical indexes of U.rhynchophylla.The results showed that the relatively low elevation planting area played a vital catalytic role on the morphogenesis,material accumulation and physiological metabolism of U.rhynchophylla.To sum up,the forest land with altitude of about 825 m,fertile and partial acid soil was more suitable for the growth of U.rhynchophylla.The ecological conditions of Danzhai County in Guizhou province are relatively suitable for growth and development of U.rhynchophylla.
