U. rhynchophylla (Miq.)Miq. ex Havil. and U. hirsuta Havil. were Rubiaceae genus Uncaria, which branch with hamulus has been embodied in Pharmacopoeia of People’s Republic of China (2010edition). Alkaloid were their effective components. Content determination and fingerprint of Uncaria has not been embodied in Pharmacopoeia of People’s Republic of China (2010edition), so it can not controlled quality of Uncaria. System research of Uncaria was lack. U. rhynchophylla (Miq.)Miq. ex Havil. and U. hirsuta Havil. from different places, different parts and different collecting times were determined with rhynchophylline content and fingerprint by the methods we builded, which were used to analysis, evaluation and control the quality of Uncaria, and provided a reference to develop and utilize of Uncaria. Research achievements as follows:1. Content detamination and Fingerprint determination of HPLC was builded.2. There was great differences between different varieties and places.11batches in26of different places of U. rhynchophylla (Miq.)Miq. ex Havil. contained rhynchophylline, which account for42.3%, content between0.014%~0.386%, and there were not detected of rhynchophylline in U. hirsuta Havil. from9different places, which account for100%.3. There was some differences on different parys in U. rhynchophylla (Miq.)Miq. ex Havil. which contained rhynchophylline.Except the leaves from Rong county(No.17) and Lipu county(No.18) were not detected rhynchophylline, other parts from different places can detected. Besides leaves has lower content of rhynchophylline, which content was0.023%, the other parts were similar(branch with hamulus was0.141%, hamulus without branch was0.139%, branch without hamulus was0.138%and stem was0.179%). 4. The content of rhynchophylline in different collecting times had great differences, which were0.137%~0.386%in a year. Among them, February was highest, which content was0.386%, the second was March, which content was0.371%, and July was the lowest, which content was0.137%.5. The standard chromatographic fingerprint of U. rhynchophylla (Miq.)Miq. ex Havil. and U. hirsuta Havil. were created, with the camptothecin peak as the external reference peak. The samples from different places were determined. It showed that the consistency of the fingerprints from different palces were good. Main chemical components which contents were high had little differences, most of them were similar, and the quality was stable.
