In order to enhance the production performance of Gushi chicken and select the best combination of quality broilers that meet the appearance characteristics of Gushi chickens,used the Gushi chicken as a male parent and the two-line matching cross combinations AH,DH and MH as the female parent to carry out a hybridized combination experiment.Three hybrid combinations were obtained in the experiment,named GAH,GDH and GMH combinations.Observing the feather and shank color,measuring and comparing the body measurement traits,growth performance,meat production and meat quality of the three hybrid combinations.Meanwhile,using the real-time fluorescence quantitative PCR to determine the expression level of ASB15,TGF-?2 and LPL gene in breast muscle,exploring the effect of expression of ASB15,TGF-?2 and LPL in muscle development and IMF content of breast muscle.The results of experiment were:1.the appearance feature in three hybrid combinations of Gushi chickenThe nascent chicken of three hybrid combinations were mainly jute and yellow feather,with a certain percentage of black feather.On week 4,the black feather disappeared,the percentage of jute and yellow feather,basically,remained the same,a certain proportion of hemp feather increased.Around the 8th week,the main color of feather had little change.The 16-week-old hens having being mainly jute and yellow feather,the 16-week-old cock had reddish yellow or red feather and well-developed crest.The shank color of nascent chicken was yellow in the three hybrid combinations,with the increase of weeks some individual turn into bluish yellow and willow shank.The shank color was generally from shallow to deep and the 16 th week was mainly willow-shank.Comprehensively,The 16-week-old experimental chickens' feather and shank color basically meets the macroscopic features of Gushi chickens.2.the growth performance in three hybrid combinations of Gushi chickenThe birth weight of GMH line was significantly higher than that of GAH and GDH line(P<0.05).2 to 16 weeks old GAH line average weight was biggest and the mean weight of 8-to 16-week-old GAH line was significantly higher than that in the other two lines(P<0.05).The relative growth rates of three combinations was a slow down as it growing.The most significant decline was observed in the cocks from 0 to 8 weeks old and the hens showed the most significant decline from 0 to 6 weeks old.GAH line had the lowest feed conversion ratio in the three lines and the feed conversion ratio of cocks was lower than that of hens.Comparing the body measurement traits in three lines,GAH and GMH lines develpoed better than GDH line.The chest width,chest angle,keel length,body length,and shank circumference of 16-week-old GAH line were significantly greater than those in the GDH and GMH lines(P<0.05).In general,the growth and body size of the GAH line were significantly higher than the other two lines during the entire feeding period.3.the slaughter performance and meat quality in three hybrid combinations of Gushi chicken16 weeks old cocks and hens of three combinations as quality chickens had good meat performance of which carcass rate were more than 85% and eviscerated yield were more than 65%.The eviscerated yield and breast muscle ratio of GAH cocks was significantly higher than that in the other two lines(P<0.05).The leg muscle ratio of GAH and GDH hens were significantly higher than that in the GMH lines(P<0.05).The abdominal fat percentage of GAH was significantly higher than that in the other two lines(P<0.05).The sebum thickness of GAH was significantly higher than that in the other two groups(P<0.05).The intramuscular fat width of GAH and GDH were significantly higher than that in the GMH lines(P<0.05).The body fat deposition in the GAH was relatively high.The leg muscle and cocks breast muscle p H were 6.0~6.5 after 45 min of slaughter that met the standard of quality chickens.The shear force of the chest and leg muscles in the GDH line was relatively large.The dry matter and intramuscular fat of GDH breast muscle had a relatively high content in the three hybrid combinations and the dry matter and intramuscular fat of GAH leg muscle had a relatively high content.The dry matter content of GDH and GMH were significantly higher than that in the GAH lines(P<0.05).The dry matter content of GAH was significantly higher than that in the other two groups(P<0.05).The intramuscular fat content of GDH cocks breast muscle was significantly higher than that in the other two groups(P<0.05).The intramuscular fat content of GAH cocks leg muscle was significantly higher than that in the other two groups(P<0.05).In comparison,the GAH group had the highest meat value.4.the expression of muscle development and fat metabolism related genes in three hybrid combinations of Gushi chickenThere was no significant difference in the expression of ASB15 genes in three combinations of breast muscles and there was no significant correlation between gene expression and breast weight(P>0.05).The expression of TGF-?2 gene in different sex of breast muscle had a inconsistent expression trends.The expression of TGF-?2 in GMH cocks breast muscle line significantly higher than that in the other two lines(P<0.05).The expression of TGF-?2 in GDH and GAH hens breast muscle were significantly higher than that in the GMH line(P<0.05).Both male and female,there was no significant correlation between TGF-?2 expression and breast muscle weight(P>0.05).The expression of LPL in GDH cocks breast muscle was significantly higher than that in the GMH line(P<0.05).The expression of LPL in GMH hens breast muscle was significantly higher than that in the other two lines(P<0.05)and the expression of LPL in GDH hens breast muscle was significantly higher than that in the GAH lines(P<0.05).In general,the expression of LPL in breast muscle was similar to that of pectoralis muscle,but there was no significant correlation.
