Estrus Induction Effect And Differentially Expressed Related Genes At Anestrous Season In Small-tailed Han Sheep

Abstract/Summary:
Small-tailed Han sheep is perennial oestrus animal,however,there was certain seasonal reproduction,especially in spring,postpartum ewes oestrus interval was longer than autumn,which greatly reduces the female fecundity,and affects the economic benefits.Therefore,the effect of estrus induction and differentially expressed related genes of Small-tailed Han sheep in anestrous season were studied.126 Small-tailed Han sheep estrus weaned at 2 months postpartum were selected as experimental animals.According to the origin of progesterone suppository for estrus induction,they were divided into three groups: New Zealand group,Australia group and domestic group,each of which was treated with progesterone suppository.According to the different methods of estrus treatment,they were divided into PRID group and PRID+PMSG group,the effects of different methods on oestrus induction in estrus were compared,and the mRNA of follicle stimulating hormone receptor(FSHR),luteinizing hormone receptor(LHR),estrogen receptor(ESR)and progesterone receptor(PGR)and progesterone receptor protein levels of ovary,oviduct and uterus of estrous and non-estrous ewes were further analyzed by real-time quantitative PCR and progesterone receptor for Western blot.The results showed that:1.The results of estrus induction showed that the oestrus proportion and conception rate of New Zealand PRID+PMSG was 93.75% and 100%,the effect of estrus was best,but the highest cost of treatment was 21 yuan per sheep.The estrus rate and conception rate of estrus induced by domestic PRID for 14 days were 85.19% and 86.96%,respectively.The effect of estrus was better,simple operation,time saving and labor saving,and the processing cost was the lowest(8 yuan).Considering the economic cost and the effect of oestrous conception,domestic PRID was the most economical and suitable for estrus induction in estrus.2.The expression of FSHR,LHR,ESR and PGR genes in ovaries,oviduct and uterus of estrous and non-estrous ewes were observed.There was no significant difference between the mRNA expression levels of LHR,ESR and PGR genes in ovaries,oviduct and uterus of estrous and non-estrous ewes(P>0.05),but FSHR mRNA in estrous ewes was significantly lower than that in non-estrous ewes(P<0.05).It is suggested that the high expression of FSHR mRNA in uterus may be related to hormone induced treatment of ewes.3.The results of traditional Western blot and automatic protein analysis system showed that there was no significant difference in the expression of PGR protein between estrus and non-estrous ewes in the reproductive organs(P>0.05).
