| Jining gary goat is a species of lamb leathers in China, characterized by precosity and high productivity. The initial mating of female goats happened approximately 6 months old.The average yean percentage of primiparity is 163.1, general speaking,one year two fetus or two years three fetus,average yean rate is 293.7%.FSH and LH is most important hormone in pituitary-ovary axis.We research the secretion changes of FSH and LH and mechanism of action by RIA and immunohistochemistry technology, approach endocrine secretion mechanism of high production in Jining gray goat, which provide theory foundation in elevating goat production and utilizating economic value.Experiment 1 Relation between pituitary contents and peripheral plasma concentrations of gonadotropins in Jining gray goatThe contents of gonadotropin in pituitary and concentrations in plasma were measured by RIA. All goats were divided into 5 groups , according to the FSH concentrations in plasma. The results suggested that the FSH contents in pituitary keep in a invariable level , There was no significant correlation (R=-0.280,P>0.1) between FSH pituitary content and plasma concentration. The change tendency of LH contents in pituitary and concentration in plasma was uniformity on the whole , their relation was significant positive correlation (R=0.544,P<0.001) . It is thus clear that there was a great difference in synthesis and secretion of FSH and LH .Experiment 2 Cellular type of pituitary gonadotropes and their changes during different reproduction phases in Jining gray goatJining gray pituitaries,collected in estrus, dioestrus and anestrus, were used to identify the cellular type of LH and FSH gonadotropes and their changes during the three phase by means of single immunohistochemistry labeling and double immunohistochemistry labeling. The result suggested difference of FSH cell and LH cell positive percentage among three phases is extreme significant (P<0.01), FSH and LH can exist simultaneously in a same GTH cell, mono FSH and LH cell positive percentage is almost no change in estrus, dioestrus and anestrus (P>0.05), FSH/LH cell shows a extreme significant (P<0.01) change among estrus, dioestrus and anestrus.Experiment 3 Ultrastructural observation of gonadotrophs in estrus and anestrus of Jining gray goatJining gray goat pituitaries, collected in estrus and anestrus, were used to observate gonadotrophs ultrastructure . The results suggested that gonadotrophs often locate in surrounding of the sinusoid blood capillary, gonadotrophs are round in shape, elliptic secretory granules are about 150~250 nm in length.In estrus, a few secretory granules locate in surrounding of the nucleus in cytoplasm, the gonadotrophs are in the state of productive secretory. In anestrus, numerous secretory granules intensively distribute in the amphi-cell.Experiment 4 Immunohistochemistry research of FSH receptor and LH receptor in ovary of Jining gray goatJining gray goat ovaies,collected in estrus,were used to identify the localization of FSHR and LHR and their changes in different follicle stage by means of immunohistochemistry and image analysis.The result suggested that FSHR only exsited in granulosa cell and cumulus oophorus and was found in all follicle. LHR mainly exsited in granulosa cell and theca interna cell. LHR first appeared in secondary follicle, LHR staining intensity is gradually reinforcement with follicle development, and there was most staining intensity in mature follicle. The amount decreased accompanied with follicle atresia. In antral follicle, FSHR cell amount gradually decreased and LHR cell increased accompanied by follicle development. |
