| Lonicera edulisTurcz.is a kind of precious wild berry with great development prospect.The fruit is too small to harvest and cannot be stored and transported,the utilization rate of wild resources was extremely low.In this experiment,wildLonicera edulis Turcz.superiorindividual in Changbai Mountain as test materials,colchicine as inducer.Tetraploid plants were induced by diploid plants,and tetraploid plants were used as materials to induce octoploid plants in the same way,three octaploid plants were successfully obtained.This experiment improved the polyploidy induction technology system of Lonicera edulis,provided theoretical basis for polyploidy induction and identification of Lonicera edulisTurcz.,and promoted the large-scale production of polyploidy superior plants.The results were as follows:1.With colchicine as inducers,adopt the Lonicera edulis Turcz polyploid induction method,induction of tetraploid or octaploid,the half lethal dose is consistent,colchicine concentration is 500 mg·L-1,the processed time for 14 days.The results of chromosome counting showed that the effect of the semi-lethal dose on the induction effect was not obvious.2.In the material treated with colchicine,the chromosomes of four materials were not doubled,and the chromosome was still tetraploid.The three materials have been doubled to octoploid,with the treatment concentration of 300 mg·L-1 and 500 mg·L-1,the processing time is 14 days,and the octaploid induction rate is 42.86%.3.Different ploidy plants’ guard cells have significan t differences in length and width.High-ploidy plants have longer and wider guard cells,but the stomatal density are in the opposite.Low-ploidy plants have smaller stomatal density.The chloroplasts in guard cells of octoploid plants was significantly higher than that of diploid plants,the chloroplast granules of octoploid plants increased significantly and color increased.Therefore,the anatomical structure of leaf can be used as a basis for preliminary identification in the polyploid induction of Lonicera edulis Turcz,Although the number of chloroplasts in the diploid and eight ploidy plant guard cells were not significantly different from the tetraploid plants,the tetraploid plants were between diploid and octoploid.There is a significant positive correlation between the chloroplast number and ploidy in the leaves of Lonicera edulis Turcz.4.The height,stem diameter,internode length and leaf length and width of the plantlets of different ploidy were significantly different.With the increase of ploidy,the plant height and internode length decreased gradually,while the stem diameter and leaf size increased with the ploidy.In addition,the leaves of the octadiploid plant were significantly thicker than the diploid and tetraploid leaves,and the color was darker and greener. |
