Forsythia (Forsythia suspensa (Thunb.) Vahl) is Oleaceae common medicinal plants, it's fruits and seeds used as medicine , has detoxification, elimination carbuncle Sanjie and other effects. Forsythia is one of the most commonly used traditional Chinese medicine,has a long history and is widely used in clinical practice. Polyploid plants have huge features, induced polyploid plants can increase the leaves, fruits and root growth of Forsythia, will has great value in medicinal and in soil and water conservation about Forsythia. In this experiment, Forsythia seedlings as materials,use of colchicine growing point of the mutagenic solution treatment method and the number of shoot-tip cell chromosome ploidy detection method as the determination identification methods to study the different colchicine concentrations and mutagenic effects of processing time. In this experiment, leaf thickness, chlorophyll content, leaf water content and conductivity determination of membrane permeability index were compared between diploid and Tetraploid plants, and to make an basic study for the further research of tetraploid Forsythia.The main results are as follows:1.The induced effects of forsythia seedlings tip In the colchicine treat forsythia seedlings growing point, we study the different concentrations of colchicine and the best combination of processing time. From the mutation effect, the 0.1%~0.3% of colchicine on the induction of forsythia seedlings growing point is valid, the concentration of colchicine of 0.1% in processing time is 48h, the highest mutation rate was 40%; concentration of 0.2 % colchicine treatment time with the growth and mutation rate, mutation rate in the 72h treatment time reached the highest 58.3%; when colchicine concentration of 0.3%, with the processing time, the mutation rate had dropped, the processing time The highest rate for the 24h variation was 56.7%, processing time for the lowest rate of 72h variation was 23.3%. In the nine treatment combinations of 0.2% treatment was 72 h, mutant highest at 58.3%, for the best combination. 2.The morphology structural changes in variant plantBasing on the morphological and anatomic study,The plants showed the general Characteristics of tetraploids,such as,thicker leaf,smaller leaf index,larger guard cells,less stomata per unit area and more chloroplasts in guard cells;The width of the palisade tissue and the sponge tissue increase,The cells of the stem point meristematic tissue level correspondingly are larger.3. identification results of the variant plantsAccording to the cytological study,the chromosome number in the polyploid plants are 2n=2x=28,but in the mutants are 2n=4x=56;Determination the DNA comparative content of the cell by cytomte the result showed that the DNA content in the cells of the polyploid plant increased one time,turned 4n by 2n .But there are also a large number of variant plants are chimera, chromosome numbers from 2n to 4n, and including a small number of octoploid cells.4. The comparison of some features of aploid and diploid ForsythiaThe results showed that:Tetraploid Forsythia plants leaf obtained significantly higher than that of diploid Forsythia plants leaf thickness;Chlorophyll content in tetraploid Forsythia was significantly higher than diploid Forsythia leaves; Tetraploid Forsythia and diploid Forsythia leaf water content in leaves was not significant.Tetraploid Forsythia conductivity and electrolyte leakage were significantly lower than the diploid conductivity and electrolyte leakage.
