Study On Technology For The Guidelines For The Conduct Of Tests For DUS Of Elderberry

The genus Sambucus is a species of characteristic plant in China.It is widely distributed in the north and south of China,and it belongs to Caprifoliaceae.Most of it is trees or shrubs and a rare herb.There are about 20 species grow in the temperate and subtropical regions of this genus,of which there are about 6 species in China.The uses and development prospects of Sambucus are extensive,with high medicinal,edible,ornamental and ecological values.With the in-depth development of the protection of the new elder species,and the increasing demand for elder trees seedlings in the landscaping,the guidelines for the conduct of tests for DUS of Elderberry is developed.The guidelines can be used for the selection of new Sambucus species.The Sambucus genus is the fifth batch of protected genus(species)in the forest protection plant directory.This article discusses the current status of protection of new varieties and DUS testing technology at home and abroad,and combines the general situation of Sambucus to explore the technology of Sambucus' s DUS test.The main research contents and results are as follows:(1)Review research.The research background,research significance,research content,work plan,materials and methods of the test guideline for new varieties of Sambucus are reviewed.By referring to the related literature and website of Elderberry,we introduced the general situation of Sambucus in China.The theoretical contents of the protection of new plant varieties at home and abroad and the development of DUS testing were analyzed.The differences between the UPOV 1987 text and the 1991 text were compared.The positive and negative effects of China's participation in the UPOV 1991 text were discussed.The response to China ' s accession to the UPOV 1991 text was discussed.And prospects for the use of newtechnologies such as molecular markers in variety testing.(2)A working plan for testing guidelines for new elderberry species in China has been formulated.Completed research on Jinan Quancheng Park,Shandong Provincial Academy of Forestry Experimental Nursery,Jinan Pingyin Elderberry Base,Binzhou Bohua Agricultural Lvyi Elderberry Base,Beijing Botanical Garden,Guizhou and other places,and formulated the traits of Sambucus genus.Observing the table,two species of growth cycle were observed and observed for the species,clones and family of Sambucus,and the characteristics of Sambucus plants were recorded.The data was supplemented in time,and the characteristics table was revised and improved.During the actual testing process,in order to avoid the interference of personality characteristics of common elderberry on commonalities,image data and data that can explain the specificity of elderberry are reserved,which prepares for the preparation of later testing guidelines.(3)Establishment of DUS Test Guideline for New Sambucus Variety.Based on the field survey results and other testing guidelines as a guideline,combined with the characteristics of the genus Sambucus,the selection of standard varieties in the DUS test technology was discussed under the conditions of the relevant regulations of the International Union for the Protection of New Plant Varieties(UPOV).As for the determination of the test traits,a traits table and a technical questionnaire for the Sambucus genus DUS test were developed.A total of 7 standard cultivars and 46 test traits(15 qualitative traits QL,13 quantitative trait QNs,and 18 pseudo-quality traits PQs)were selected for the DUS test of the new Sambucus cultivars.18 traits were measured and included in the technical questionnaire.There were 28 other traits.The test guidelines specify the selection of test materials,test methods and observation methods for different traits,and some of the traits are illustrated.After the completion of the above work,we communicated and communicated with many domestic experts and revised it,and finally completed the test guide for the new species of Sambucus in China..(4)A database of known species of the Sambucus was established.Reference traits and their classification criteria,combined with other types of information,toestablish a database of known genus Sambucus,to make the construction of DUS test system of new Sambucus cultivars more complete.
