Lychee stink bugs,Tessaratoma papillosa(Drury)are destructive pests of litchi and Longan,they can cause great impact on the growth and production.This study focuses on symbiotic bacteria of litchi bugs in variety,bacterial differences between provinces as well as the advantage bacterial species and so on.First,different sampling areas are analyzed of litchi bugs between adult and nymph about the diversity of the symbiotic bacteria,then see province as the basic unit,to research that whether there is significant difference among the samples that belong to different provinces symbionts;in the end,the two stages of insects--adult and nymph are studied to analyze whether there is significant difference in the symbiotic bacteria between different species,and the main results of this study are as follows.1.Analysis of the diversity of symbiotic bacteria in samples from different sampling areas by 16S rRNA V3+V4 region high-throughput sequencing.Based on 16S rRNA V3+V4 high-throughput sequencing,We analyses the diversity of symbionts species from sampling areas,the results shows that different area samples' advantage bacteria is Proteobacteria in phylum,and in genus it is belong to the Enterobacteriaceae-unclassified;Actinobacteria is the dominant bacteria of different samples in different sampling regions,and Streptomyces is the dominant bacteria in genus.Firmicutes and Bacteroidetes in the two stages of the insect is not high,both of them contain a small amount of bacteria;in the two insect' stages,all samples contain a certain amount of Pantoea.Through the analysis of the representative OTUs in the sample,it can be concluded that the symbiotic bacteria of the bugs mainly focused on Enterobacteriaceae and Streptomyceae.2.The difference of symbiotic bacteria between different provinces is analyzed.Based on the analysis of the diversity of samples in adult and nymph in different provinces,we mainly research whether there is a significant difference between nymph and adult in different provinces.Among adult,the P value of a diversity index is among 0.0865(ACE)-0.3335(Goods coverage),which is greater than 0.05;Among nymph,the P value of ?diversity index is among 0.096(shannon)-0.754(ACE),which is greater than 0.05,too.The results shows that in a there is no significant difference.Among adult and nymph has the same results.Among adult,the P value of ? diversity index is 0.224,Among nymph it is 0.553.The results shows that in ? there is no significant difference.Among adult and nymph has the same results.It can be concluded that geographical location or environmental factors have little effect on the composition of symbiotic bacteria.3.Analyzing whether there is significant difference between the two stages of insect in adult and nymph.Three provinces are selected to study the composition and proportion of the symbiotic bacteria between nymph and adult of samples in each province,the diversity of the species,the number of the species,alpha and beta index.The results shows that the symbiotic bacteria of nymph in each province is different from that of adult.In phylum,the adult's dominant symbiotic bacteria is Proteobacteria,and the nymph is Actinobacteria.In genus,the dominant symbiotic bacteria of the adult is Enterobacteriaceae-unclassified,the nymph is Streptomyces,further on variance analysis of the adult and nymph of various bacteria percentage,the results shows the P value of the bacteria that have little percentage among 0.135(Fujian Province)-0.713(Guangxi Province)in phylum and in genus among 0.396(Guangdong Province)-0.412(Guangxi Province)The results shows that low percentage bacteria between adult and nymph is no significant difference.As for Dominant and subdominant symbionts,P value among 0-0.04 in phylum and in genus among 0-0.05,this results shows that there is significant difference between adult and nymph.As for alpha and beta diversity,the P value of shanon,simpson,and ? in Guangdong province and Guangxi province among 0.012-0.043.which shows that the adult and nymph of Guangdong province and Guangdong province have significant difference,however,due to the adult and nymph do not constitute a significant difference in Fujian province,so the data is not sufficient to illustrate that the two insect stage have significant differences in alpha and beta diversity.
