Hydroperoxides formed in the catalytic process of lipoxygenase are important intermediates in the pharmaceutical,chemical,food,and other industries,and have attracted the attention of researchers.The study found that lipoxygenase has high activity in legumes,of which the highest activity in soybean,but because of the uneven content of lipoxygenase in different varieties and easy to degenerate,it restricts its application and development.This study explored the changes of soybean lipoxygenase activity from both resource and ecology,laying a foundation for the cultivation of new soybean varieties and the development of soybean production.Through the evaluation of soybean variety resources,the level of soybean breeding in China can be improved,and the depth and breadth of research and utilization of soybean variety resources can be further strengthened.Therefore,it is an important basic research work to study the lipoxygenase activity in soybean seeds.This study mainly focuses on two aspects.One is the soybean varieties(lines)in the northeastern region.The activities of LOX-1,LOX-2,and LOX-3 in different varieties of soybean are determined to analyze changes in lipoxygenase activity.Regularity and application of statistical analysis software and simple linear analysis of the correlation between soybean lipoxygenase activity and soybean oil and protein content,and their resource evaluation;Second,the impact of different environments on lipoxygenase activity was studied from the ecological perspective;The main results of this study are as follows:1)Using 311 soybean varieties(lines)from Northeast Agricultural University's Xiangyang Experimental Base as materials,it was found that there was a significant difference in the activity of lipoxygenase among the varieties,and the main peak of the frequency/group distribution curve of the variety was obvious.The activity of LOX-1 changed the most,with a minimum of 1.474×103U/ml,a maximum of 40.388×10~3 U/ml,a mean of 20.148×10~3 U/ml,and a coefficient of variation of 34.82%.LOX-2 The activity range of activity was 0.147×10~3 U/ml~1.112×10~3 U/ml,mean value was 0.524×10~3 U/ml,coefficient of variation was 35.39%;LOX-3 activity range was 0.371×10~3 U/ml~1.777×10~3 U/ml,mean value 0.990×10~3 U/ml,coefficient of variation 23.05%.The correlation analysis showed that there was a significant positive correlation between LOX-1 and LOX-2 and LOX-3,and there was no significant correlation between LOX-2 and LOX-3.The cluster analysis divided 311 soybean varieties classified into three cultivars,about 43.41%of the soybean varieties belonged to the low-lipoxygenase-active cultivar group,and about 44.05%of the soybean cultivars belonged to the middle-lipoxygenase-active cultivar group;about 12.54%.The soybean varieties belong to the high lipoxygenase active variety group.By analyzing the relationship between soybean lipoxygenase activity and oil and protein content,it was found that there was a significant correlation between soybean oil and protein content and lipoxygenase activity.There was a significant positive correlation between oil content and LOX enzyme activity,and there was a significant negative correlation between protein content and LOX enzyme activity.2)The soybean lipoxygenase(LOX)activity was measured and the results were analyzed by the variance analysis of the effect of six soybean varieties(lines)in the experimental area of Northeast Agricultural University.The results showed that the environment,genotypes and their interactions had a significant effect on the activity of lipoxygenase.Among them,genotype was the main factor affecting the activity of soybean lipoxygenase,and the environmental effect was the least.Comparison between sites,the average value of soybean lipoxygenase activity in the Xiangyang pilot was a minimum of 25.761×10~3 U/ml,and the average value of soybean lipoxygenase activity in the Hulan pilot was a maximum of 31.760×10~3 U/ml.The average value of enzyme activity was 28.036 x 10~3 U/ml between the two.
