Notopterygium H.Boiss is the main source of traditional Chinese herbal medicine,From the book Chinese Pharmacopoeia,the Notopterygium medicinal materials is the dry root of Notopterygium incisum Ting ex HT Chang and Notopterygium forbesii H.de Boiss.Because of its wildly use,a larger market was demanded.However,under the natural conditions,the germination rate of Notopterygium seed was very low.While the seeds of Notopterygium were difficult to germinate in natural conditions,our purpose of the research is to understand its specific reasons.From relevant studies,we learned that,as a method of man-made control of the environmental conditions,Stratification can raise the Speed of the ripening process,and shortening the time for the seeds to complete the ripening,so that we can finish our study as success as possible.During the stratification,we sampling every period of time,in order to know the development of embryo of Notopterygium seed in the process of after-ripening preliminary,we take the paraffin slice,and combined with the changes of related physiological date during the process of after-ripening.In our experiment,the seeds of Notopterygium collect from HuiNing County,Gansu Province,were treated for a period of 200d,and the relevant indexes were measured periodically during the stratification process.In this experiment,it is the first time to explore the late-maturing process of the seed of N.forbesii Boiss.combine with the embryo changes and physiological indexes in the after-ripening process,and finally the following results were obtained:1.The develop of embryo:In the early time of the after-ripening,about0?40day's stratification,we observed a "bright spot" in the seed,as time goes by,the "bright spot"expanded,at the same time,the embryo division and differentiation.After 100-120 day's stratification,we can observe the embryo directly,at this time,the "bright spot" become an intervening space,the embryo stay in the intervening space freely,and mature gradually.Finally,the radicle breakthrough the seed coat and the seed germinated.2.Storage material:The seed germination in the early stage(0?100d)of the stratification,the quality of the seed of N.forbesii Boiss will decrease rapidly,later(100d?200d),it become smooth;the content of protein changed a lot during the stratification,in 0?40d,it changes a little,and during 60?80d,it decreases specially,in 100?200d,it increases and stay in a high level;the content of starch is in a very low level in the N.forbesii Boiss seed,and it changed not that obviously,after decreases in 0?20d,it changes a little in 20?180d,finally it increase a lot in 180?200d;Lipids changes almost in 0?40d,the content of lipids decrease from 34.1%to 12.6%,then it becomes steady in 40?200d.3.Antioxidase:As time goes by,the activity of CAT increased,during 0?140d,the activity of CAT increases not that obvious,but in the time of 160?200d,it changes obviously and fluctuating;the activity of SOD expressed as increase in 0?40d,then decreased in 40?80d,finally,it is increase to a high level again during 80?200d.4.Glucose metabolism enzymes:The activity of Alpha-amylase increase in general,in 0?160d,it is stable,only change in60?100d,during 160?200,the activity of Alpha-amylase increase rapidly;the activity of MDH is decreased as the seed become mature,in contrast to the activity of G-6-PDH.As the seed become mature,the activity of MDH decrease but increase in 40?80d,then in 80?200d,it decrease rapidly and become stable in the rest of the time,and the activity of G-6-PDH perform increase at last,in 0?160,it changes fluctuantly,then it increase in 160?200d.
