The secondary metabolites and their biological activities of entomogenous fungi Paecilolyces gunnii and edible-medicinal fungi Morehella esculenta were explored in this thesis.Paecilomyces gunnii was cultured in the potato dextrose medium with a total volume of 60 L by liquid submerge fermentation.Fermented broth was removed the mycelium and then extracted with ethyl acetate.The organic layer was dried over Na2SO4(anhydrous)and concentrated in vacuo to afford 6.3 g organic crude.The crude was purified by repeated column chromatography(RP-18,Sephadex LH-20,and silica gel)to afford five compounds(xp05,xp06,xp07,xp09,xp 15).Their structures were elucidated through NMR,MS,UV,IR,X-ray single crystal diffraction and other physicochemical data.xp05 was a novel macrocyclic tetralactams and named as gunnilactam A.xp06,xp07,xp09,and xp15 were macrodiolides.xp06 was a new macrodiolide and was named as gunnilide A.xp07 was identified as colletallol.xp15 was a mixture of two analogues,one of which was colletol and the other was a new derivative of colletol and was named as colletol B.xp09 was a new analogue of bis-osumundalactone and named as bis-osumundalactone B.xp081 was obtained from the alkaline hydrolysis of xp05 and was identified as 7-amino-4-hydroxyoct-2-enoic acid which was a new none protein amino acid and was named as gunnine A.xp081 showed inhibitory effect on the growth of bacillus subtilis with MIC of 0.87 mM.Compounds xp06 and xp09 possessed the antioxidant activity by the method of TLC-bio-autography.Morehell esculenta was cultured in the potato dextrose medium with a total volume of 40 L by liquid submerge fermentation,the fermentation time was 15 days at 28?.The dried weight of mycelium was 300.0 g.150.0 g of the dried mycelium was extracted with methanol.The methanol extract was concentrated in vacuo to afford 5.6 g organic crude.The fermented broth was extracted with with ethyl acetate.The ethyl acetate layer was concentrated in vacuo and afforded 3.4 g organic crude.The organic crude was subjected to column chromatography over RP-18,Sephadex LH-20,and silica gel.Seven compounds(xx01,xx02,xx04,xx05,xx07,xxl,and xx14)were isolated from the fermentation broth.Two compounds xx08 and xx15 were isolated from the mycelium.These compounds were elucidated by NMR,MS,X-ray single crystal diffraction.Compounds xx04,xx07 and xx14 were new natural products and was named as 3,4,5,7-tetrahydroxyisochroman-1-one,2-hydroxy-6-methylnicotinaldehyde,and 4-hydroxy-1-(3-hydroxyphenyl)butan-l-one,respectively.Compunds xx015 xx023 xx05,xx085 xx11,and xx15 were known compounds and was identified as 2-phenylacetic acid,benzoic acid,4-(hydroxymethyl)-benzoic acid,4-hydroxy-nonenoic acid,3,5-dihydroxy-4-methoxy-2-methylbenzoic acid,and 1,2,4-Benzenetriol,respectively.It was indicated that compounds xx04,xx05,xx07,and xx11 possessed the high antioxidant activities by the method of TLC-bio-autography.It was concluded that Paecilomyces gunnii and Morehella esculenta could produce the aboundant secondary metabolites with structural diversity and bioactivities.This paper will lay a foundation for development of their pharmacological potential.
