| Fomes is a large ftuigus.Fomes fomentarius,a Basidiomycete,Polyporales fungus belonging to the genus of Fomes.Fuscoporia punctata(Fr.)Cunn belongs to Polyporaceae),Fuscoporia.They all grow on broad-leaved trees and have a variety of medicinal activities.he fruiting bodies of Fomes fomentarius are used as medicines to treat cancer diseases such as esophageal cancer.Fuscoporia punctata(Fr.)Cunn has been reported by modern pharmacology to treat arrhythmia.However,there are few reports on the chemical constituents and biological activities of the fruit bodies of Fuscoporia punctata(Fr.)Cunn.In order to further understand the pharmacodynamic substance basis of these two macrofungi.Therefore,this study investigated the chemical constituents and biological activity of these two large fungi and looking for novel structure and good biological activities of compounds in two large fungi to provides a theoretical basis for the discovery of leading molecules in large medicinal fungi,the creation of new drugs and the development of products by utilizing their resources.The compounds were isolated and purified by silica gel,ODS chromatography Sephadex LH-20 column chromatographies and preparative HPLC method.The structures of the compounds were identified through the combined analysis of physicochemical properties and NMR spectrum data.Eleven compounds were isolated from 95%ethanol extracts of the fruit bodies of Fomes fomentarius,2,4-dihydroxy-3,5-dimethylacetophenone(1),3,4-dihydroxybenzoic acid ethyl ester(2),4-(4-hydroxyphenyl)but-3-en-2-one(3),ergosta-7,22-diene-3p-ol(4),ergosta-7,22-diene-3-one(5),1,5-bis-(3,4-dihydroxyphenyl)-1,4-pentadien-3-one(6),3,4-dihydroxyacetophenone(7),p-hydroxybenzaldehyde(8),4-hydroxyacetophenone(9),3,4-dihydroxybenzaldehyde(10)4-(3,4-dihydroxyphenyl)-3E-buten-2-one(11).Compound 6 was isolated from Fomes fomentarius for the first time.Fourteen compounds were isolated from 95%ethanol extracts of the fruit bodies of Fuscoporia punctate:3,4-dihydroxybenzaldehyde(12)、friedelin(13)、inonotusin B(14)、inoscavin C(15)、phellibaumin A(16)、phellifuropyranone A(17)、inoscavin A(18)、phelligridin D(19)、phellibaumin D(20)、catechol(21)、p-hydroxybenzoate(22)、pyrogallol(23)、p-hydroxybenzylideneacetone(24)、1,6-dihydrocyclobuta[1,2-b:4,3-b’]dipyrrole-2,5-dione(25).All compounds were isolated from Fuscoporia punctate for the first time,compound 25 is new compound.The compounds was used to evaluate the biological activities.The a-glucosidase inhibitory activity of compounds was evaluated by PNPG method,the anti-oxidative ability of the fruiting bodies of Fomes fomentarius were studied by DPPH method,Similarly,96-well plate microanalysis were used to determine the antibacterial activity.Some of the compounds were measured the protein tyrosine phosphatase(PTP-1B)inhibitory activity.by PNPP.In vitro activity screening result showed that compounds 6,7 and 10 had inhibitory activity against a-glucosidase;compounds 1-11 showed anti-oxidative ability,Compounds 7 has a high anti-oxidative ability of 71.99%.compound 1,3,6,7,11 exhibited acetylcholinesterase inhibitory activity,compound 6 showed antimicrobial Sarcina lutea,Bacillus subtilis activity.The minimum inhibitory concentrations were 64 mg·L-1 and 128 mg·L-1,respectively.Compounds 14、15、17-19 exhibited higher a-glucosidase inhibitory activity,IC50 values were 256.18、127.77、463.84、264.06、122.99 μM.compounds 15-17、19、20 showed anti-oxidative ability;IC50 values were 30.45、227.82、32.90、25.79、6.71 compounds 14、15、17-20 exhibited acetylcholinesterase inhibitory activity;The acetylcholinesterase inhibitory activity of Compounds 15 is 32.41%.compounds 14-16、19、20 had inhibitory activity against protein tyrosine phosphatase(PTP-1B). |
