In this paper,in order to explore the effects of Eucommia leaf on sheep serum metabolomics,meat quality and fatty acid metabolism in liver,30 small-tail sheep were randomly divided into 3 groups including a control group(CTL),10% Eucommia leaf-added group(EUL1)and 20% Eucommia leaf-added group(EUL2).CTL group was fed basal diet,while 10% and 20% Eucommia leaves of dry matter basis were added into EUL1 and EUL2 respectively.The whole test lasted 105 days,including pre-feeding 15 days,formal fed 90 days.In the end,blood samples were collected.Five with similar weight from each group were selected to slaughtered.Liver samples and muscle samples were processed and analyzed.Test results are as follows:1)After adding Eucommia leaf,three groups muscle samples changed in the fat content significantly(P <0.05).there was highest fat content in EUL2 which increased 17.19%,compared with the CTL;muscle protein content increased t increased by 9.69% in EUL1,which had the highest protein content(P> 0.05).Compared with the CTL saturated fatty acid content tended to decrease in both groups,while the difference was not significant.Unsaturated fatty acids including MUFA and PUFA were significantly increased in EUL1 EUL2(P <0.05).2)In sheep blood biochemical results,glucose,glucocorticoids,high density lipoprotein,insulin,leptin,non-esterified fatty acids and very low density lipoprotein in EUL1 and EUL2 has increased compared with CTL,in which EUL2 increased significantly(P <0.05).Sheep serum samples were analyzed by LC-Ms and 1570 feature data were detected.Combined with PCA,PLS-DA and OPLS-DA analysis,26 kinds characteristic metabolites of Eucommia adding in sheep serum difference were identified..3)On m RNA level,compared with the control group,CPT1 A,LPL of EUL1 were significantly increased(P <0.05)and SREBP1-c decreased significantly(P <0.05);SCD1,SREBP1-c and PPAR? transcriptional level of EUL2 significantly decreased(P <0.05).On protein level,SCD1 protein expression in EUL2 significantly reduced(P <0.05);LPL tended to express more than in the control group(P in the EUL1> 0.05).
