ObjectiveTo sublimate the method of quality evaluation of Callicarpa nudiflora,ensure the safety and effect of clinical application,and make the foundation of further development and exploitation of Callicarpa nudiflora plant.This paper evolve the quality standards of Callicarpa nudiflora and research the optimal collection period of growth places between Baisha and Lingao in Hainan province.Including literature research,morphologic histology,the medicine routine inspection including determination of water,and determination of ash,medicinal microscopic identification,TLC identification,assay and the approach of the optimal collection period.MethodsFirstly,the Callicarpa nudiflora medicinal material base of former research,plant morphology,medicinal properties,microscopic characteristics,chemical composition,quality evaluation and pharmacological action and clinical application,etc,has carried on the comprehensive review,by refer to literature information,the general situation of the research reflect the Callicarpa nudiflora,for the Callicarpa nudiflora provides a theoretical basis for further study;Combining previous research,laid on powder microscopic characteristics research,provide the basis for the crude drug identification;Then establish TLC method for qualitative identification of Luteoline and 5,4'-dihydroxy 3,7,3'-trimethoxyl of flavone as indexes of Callicarpa nudiflor,and HPLC simultaneous determination of Acteoside,Luteoline and 5,4'-dihydroxy 3,7,3'-trimethoxyl of flavone in the Callicarpa nudiflor.Finally,taking the compositions of Acteoside,Luteoline and 5,4,-dihydroxy 3,7,3'-trimethoxyl of flavone as investigation index,estimate developmental tendency of chemicalresources in different collection period and different regions of Callicarpa nudiflor in hainan,to provide the data basis of determine the optimal collection period of Callicarpa nudiflor.Results1.The petioles,main veins,lateral veins were densely costed gray-brown stellate hairs and unicellular non glandular hairs,on the surface of leaf was glabrous,the superficies inferia of leaf was costed by yellow branched hair and stellate hair,and full of yellow glandular dots by microscopic microscopy.The branched hair,stellate hair,single,multicellular nonglandular hair can be seen by the powder production.The Luteoline and 5,4'-dihydroxy 3,7,3'-trimethoxyl of flavone TLC qualitative identification method of Callicarpa nudiflor is to sink medicinal alcohol extraction of acid extraction after removing impurity of the concentrate to the point of test sample solution.In saturated methylbenzene:acetoacetate:methanoic acid?5:2:0.5?as developing agent in the reversed-phase silica gel GF254 thin layer plate,observed under uv lamp?365 nm?,with corresponding fluorescence spots.2.The HPLC determination method of Acteoside,Luteoline and 5,4'-dihydroxy 3,7,3'-trimethoxyl of flavone was applied on Agilent TC-C18?250 mm×4.6 mm,5?m?,at 40 ? under the condition of gradient elution with mobile A:methanol and mobile B:0.15%phosphate.The flow rate was 1.0 ml · min-1,the detection wavelength was set at 350 nm.The experimentmal result for the linear range of Acteoside,Luteoline and 5,4'-dihydroxy 3,7,3'-trimethoxyl of flavone was 29.6?1490.0?g ·mL-1?r=1.0000?,1.0?50.0?g·mL-1?r=1.0000?,2.6?130.0 ?g·mL-1?r=1.0000?,the average recovery was 98.81%,99.38%and 100.79%,RSD was 1.49%,1.46%and 1.17%,respectively.3.the optimal collection period about the compositions of Acteoside,Luteoline and 5,4'-dihydroxy 3,7,3'-trimethoxyl of flavone were in April,October and April,January respectively.Between different regions Acteoside in Lingao is the better,flavonoids ingredients Baisha origin is better.The dynamic accumulation regularity of main active ingredient analysis and evaluation in different regions and different collection period show that the optimal collection period of Callicarpa nudiflora were in March and April in the spring.Conclusion:In this paper about the Callicarpa nudiflora quality evaluation study,established TLC identification method and HPLC method is used to simultaneously determine the assay of multiple effective components in Callicarpa nudi flora,for further perfect medicinal materials quality control method in Callicarpa nudiflora.To have the analysis and evaluation of effective components in different collection period in different origin,summed up the dynamic accumulation regularity of effective components,preliminarily determine...
