The infection of myxospore in the throat of allogynogenetic gible carp is a serious parasitic disease which is endangering fisheries development.In the present study,we investigated and studied of myxosporeans parasitic in crucian carp in Liyang city,Jiangsu province,and established a specific and sensitive PCR test method for M.honghuensis,so as to provide some theoretical basis for life cycle of myxosporeans and prevention of myxosporidiosis.1.Investigation of myxosporeans parasitic in crucian carp.We investigated pond-reared crucian carp in Liyang city in August,2013.In this survey,we found one myxospore from throat of crucian carp.According to the morphologic characteristics and 18S rRNA genes sequence,this myxospore,was identified as M.honghuensis Liu,2011.The taxonomic and phylogenic relation of M.honghuensis was analyzed based on 18S rDNA.2.Nest-PC detection method for M.honghuensis by Genome DNA solution of M.honghuensis was diluted to 100?10-1?10-2?10-3?10-4?10-5?106?10-7 with sterile distilled water.And then nest PCR reactions were carried out using the universal eukaryotic primers ERIB1&10 and the M.honghuensis specific primer Nest-F&R.After the end of reaction,comparing with the result of one-round PCR and nest-PCR by agarose gel electrophoresis,we found that the sensitivity of nest-PCR was 102?103 time higher than the one-round PCR.3.Through field investigation of fish ponds,visiting fish medicine shops,consulting technical director of large aquatic feed company,reference and other related prevention of human parasites in ancient and modern theories,instruments and measures,imply the source of cobalt-60 radiation Wuxi El Pont radiation technology co.,ltd to radiation-attenuated the myxosporean.Radiation absorbed dose:100Gy(10krad),150Gy(15krad),the 200Gy(20krad)and injected the myxospore into the body of Carassius auratus after irradiation.There are 4 groups,including 1 control group.There are 20 crucian carps in each group.Breed 48 days after the extraction centrifuge directly in serum tube agglutination test,radiation absorbed dose 100Gy(10krad)the prescription price of 1:640,radiation absorbe dose 150Gy(15krad)prescription price of 1:1280,radiation absorbed dose 200Gy(20krad)prescription price of 1:320.Titer in the control group is zero.Radiation absorbed dose 150Gy(15krad)crucian carp when laryngeal myxosporean inhibition and destruction that has had the biggest effect.This dose caused by weak throat myxosporean immunology of crucian carp have maximum protection.The experiment for application of nuclear radiation technology industrialization in producing vaccine of disease by myxospore in the throat of allogynogenetic gible carp have made useful pioneer,exploration and attempt.
