This experiment used Taxus cuspidata branches stem tips as a material which from Helong in Jilin Province and conducted a series of tissue culture research.The main results were as follows:1.Female plant of Taxus cuspidata stem tips with four leaves and length of 15 mm-18 mm are more suitable for bud differentiation and growth.2.In different sampling time,draw the materials from March to November are suitable for bud differentiation of Taxus cuspidata,the highest bud differentiation rate of material was drawn from January.3.In the four basic culture mediums(DCR culture medium,MS culture medium,B5 culture medium,WPM culture medium),the WPM culture medium compared with other three kinds of culture medium is more suitable for bud differentiation and growth of Taxus cuspidata.4.The medium with auxin and cytokinin,the concentration ratio have influence of the bud differentiation and growth:IBA combined with low concentration of 6-BA,the bud differentiation and growth conditions are relatively,NAA combined with high concentrations of 6-BA,the bud differentiation and growth conditions are relatively.5.The medium only contains Auxin the promotion of bud differentiation and growth are better than auxin and cytokinin mixed.And in different types and concentrations of auxin,2,4-D compared with other two auxins is more suitable for bud differentiation and growth of Taxus cuspidata,the optimum concentration of 0.4mg/1-0.8mg/1.6.The activated carbon and ascorbic acid promote bud differentiation and growth of Taxus cuspidata and it is better than VC in the inhibition of callus formation and browning and bud differentiation and growth promotion,the optimum concentration in 1.0g/1-1.5g/1.7.Different PH values and sucrose concentration have some influences on bud differentiation and growth of Taxus cuspidata:PH 5.2 is more appropriate to bud differentiation and growth of Taxus cuspidata during four gradient PH value;20 g/1-40 g/1 is suited to bud differentiation and growth of Taxus cuspidata during four sucrose concentrations.8.The most suitable culture conditions for bud differentiation and growth of Taxus cuspidata:temperature around 25℃,illumination 12h/d-16h/d,light intensity 30001ux-50001ux.9.Most suitable basic medium for induced adventitious of Taxus cuspidata is DCR medium;the optimum concentration of hormones is 1.5-2.0mg/1 of IB A or 0.1-0.2mg/1 of ABA,the effect of combined with two hormones are better.
