Research On Production Of Taxol By Tissue Culture Of Taxus Chinensis Var Mairei

Taxol is a new kind of anti-cancer medicine having assured curative effect,but its nature source——Taxus are very exiguous and close to deracinate.By tissue culture of Taxus chinensis var maire,this research has optimized the culture conditions,proved content of taxol in callus,offered basal datas for protection of Taxus resource and clinical using of taxol.This research obtains following results:1. Best disinfection project for explants of Taxus chinensis var maire: Based on trial tests with single factor, Taxus chinensis var mairei's best explant screened for tissue culture are annual tender stems and by first time of orthngonal test, the most adaptable disinfection project was confirmed. It is to dip stems in 70% of ethanol for 30s, then dip them in 2% of hydrargyrum chloride for 8min.2.Optimization of inducement culture medium for callus of Taxus chinensis var maire: Fundamental culture medium was MS. Based on trial tests with single factor, orthngonal test was applied to concentrations of three hormone and ratio of the most adaptable hormone inducing callus of Taxus chinensis var mairei was confirmed, it is as following: NAA concentration, 1.5mg/L; 6-BA concentration, 0.1mg/L; 2,4-D concentration, 0.25mg/L.3.Optimization of productive culture medium for callus of Taxus chinensis var maire: Fundamental culture medium was B5. Based on trial tests with single factor, orthngonal test was applied to concentrations of three growth regulating substances and accession project of growth regulating substances for productive culture medium of passage culture of callus of Taxus chinensis var mairei was confirmed, it is as following: NAA concentration, 0.5mg/L; 6-BA concentration, 0.5mg/L; hydrolytic casein concentration, 0.25mg/L.4.Screening of anti-browning dose for callus of Taxus chinensis var maire: According to core function of PPO in browning of callus of Taxus chinensis var mairei, anti-browning substances capable to restrain PPO in various aspects were selected.
