Serum procalcitonin is a sensitive indicator for the differential diagnosis of systemic bacterial infections,and is also a key indicator of the severity of the disease.It is more sensitive and specific than traditional inflammation indicators.Antibiotics not only kill bacteria,but also have good inhibition and killing effect on pathogenic microorganisms,but the abuse of antibiotics will lead to the spread of resistant bacteria and seriously affect human health.In view of this,in 2012 China officially promulgated the “Guidelines for Clinical Use of Antibacterials” policy,strictly controlling the use of antibiotics.The special clinical significance of serum procalcitonin can effectively guide the rational use of antibiotics in clinical practice,and the breakthrough of instant test analyzer and fluorescent immunotomography technology makes the detection and clinical application of serum procalcitonin become more and more extensive.The real-time assay analyzer is based on fluorescence immunoassay.Fluorescence immunoassay is divided into time-resolved fluorescence technology,fluorescent microsphere technology,up-lighting technology and quantum dot fluorescence technology according to the different tracer tracers.This paper analyzes and studies the above various types of immune technology,and expounds the principle of fluorescent immunotechnology and its clinical application,system advantages and disadvantages.Emphasis is placed on the analysis of the principles of time-resolved fluorescence immunoassays that are currently prevalent in the market and relatively mature in technology.In order to detect the content of procalcitonin in human blood quickly and accurately,this paper designs a quantitative analysis system of fluorescent immune reagents based on fluorescence immunoassay principle,and develops a whole set of analytical systems for quantitative detection of fluorescence immunity by designing optical mechanism,studying hardware structure system and electronic circuit system.The system includes functional modules such as detection project identification,peripheral data exchange,fluorescence signal detection,information recognition,storage stack and host computer Management software.The whole process of sample quantitative detection can be controlled,eventually through human-computer interaction and printer output results,and through the storage stack to retain historical data.The design of the fluorescence Immunoassay and the accompanying procalcitonin reagent for the systematic performance verification study,analysis of the instrument to detect the performance indicators to achieve the desired purpose.The main contents of the study include repeatability,reporting range,indoor quality control,accuracy,interference experiment,precision,linear range,clinical trial comparison.After systematic research,all the above research indexes can meet the performance requirements of the medical field for the test instrument,which shows that the method of procalcitonin original detection proposed in this paper is feasible.
