Immunoassay technology is based on the principle of specific binding between antigen and antibody,and has become an important part of modern medical examination by marking the content of one substance in the antigen or antibody.Immunoassay technology not only has been widely used in the field of medical examination and clinical diagnosis,but also can be used in the field of environmental detection,food safety,drug detection and so on.Immunofluorescence analysis of time-resolved fluoroimmunoassay technology(TRFIA)is a relatively new ultramicroanalysis method,has the advantages of simple preparation,multi-marker,repeat test etc.,and the sensitivity and specificity of the test has exceeded the radioimmunoassay and enzyme-linked immunosorbent assay.Another advantage of TRFIA is that it is easy to realize the portable design with the immune chromatography technology,and it can reduce the volume of the detection reagent strip,simplify the detection and analysis module and achieve rapid detection function.TRFIA to meet the current real-time detection(POCT)characteristics required for convenient,fast and efficient,has attracted wide attention at home and abroad,the research on TRFIA analysis instrument early detection research institutions and enterprises abroad,and domestic relative fall in the detection accuracy and the function of the instrument in foreign.Based on the research of the basic principle of TRFIA,the TRFIA analyzer system is designed and developed based on embedded microprocessor technology and photoelectric sensor technology.The completed research work mainly includes:(1)Designed whole system of the TRFIA analyzer system.We had study and designed the system with the time-resolved method,and had discussed the key modules such as micro controller,ultraviolet light source and photoelectric detector.(2)Designed and developed the hardware circuit of the TRFIA analyzer.The main modules include: microcontroller,UV LED,motor drive,photoelectric gate,photoelectric sensor,signal conditioning circuit,analog-to-digital converter,FLASH power supply,storage,display and other interface circuit etc..The whole circuit system based on the embedded microprocessor STM32F103 as the control core and calculation.By using UV LED as excitation light source fluorescent material and using silicon photodiode as detector,and other necessary interfaces and peripherals,the system could detect and display the results of chromatographic test strip.(3)Developed the software system of TRFIA analyzer.The software is based on the real-time operating system FreeRTOS and HAL library development,and the use of the Keil MDK-ARM integrated development environment for the preparation of code and program download.The software of the system is running in the microcontroller STM32F103,which realizes the task of driving,scanning process control,data processing and so on.(4)Research based on the developed system platform.The study includes data processing methods such as time resolved acquisition method,peak detection method,and substrate estimation.(5)Tested the system.After testing,the instrument has a wide dynamic range and better repeatability(CV<0.2%)when the concentration greater than 100?g/mL.It has a better application prospects in Point of Care Testing.
