In recent years,with the rapid development of gene technology,the PCR technology has been widely applied in all fields of biology.PCR's temperature calibration is very important.In view of the current domestic PCR calibration work is still in the initial stage of development,this paper takes "Research on PCR Analyzer Temperature Calibration Device Based on Temperature Itinerant Detecting Instrument",to build the hardware platform of the PCR instrument temperature calibration device,to design the temperature calibration device for the PCR instrument,and to evaluate the uncertainty and formulation of the PCR temperature calibration device.It has important academic value and practical significance to trace the source and result of temperature calibration device of PCR.This paper first analyzes the background and significance of the research on the temperature calibration device of the PCR instrument.Taking the temperature calibration as the breakthrough point,this paper makes a detailed analysis of the temperature control mode of the PCR instrument,the importance of the temperature to the PCR instrument and the status of the calibration of the PCR instrument at home and abroad.The aim of the development of the PCR device is determined.The main works of this paper includes:? On the basis research and comparative analysis of the existing temperature measurement method,combined with PCR temperature high accuracy,fast rate of temperature rise etc,calibration device selects temperature measuring probe as the temperature measuring element.According to the special external type of the heating parts of the PCR analyzer and the need of measurement in closed cover condition,a special temperature sensor is designed to fix the temperature probes on a special printed circuit board.The signal is collected by multi-channel temperature detector.The device adopts Mod Bus bus protocol as the communication protocol.To the calibration work,a special software is designed for the convenience of calibration.Software is designed on VC++6.0 the development platform.The software data interface is clear,and the operation is convenient,with data display interface,channel selection,historical data playback and other functions.? Paper analyzes the factors affecting the calibration device.A class uncertainty analysis is conducted for the uncertainty caused by repeated measurement.B class uncertainty analysis is conducted with the uncertainty caused by other factors.Finally,the synthesis of standard uncertainty,calculate the unit standard uncertainty is less than 0.01?.? In order to make the calibration device go up to the source,a set of traceability plan for the calibration device is designed.The second class standard platinum resistance thermometer,FLUKE 7321 thermostaic bath and special isothermal block are used as the main standard.Comparative measurement is the main method of this temperature traceability.The temperature calibration device can be traced back to the standard equipment of the first class standard platinum resistance thermometer in NIM China.Finally,the calibration device is applied to the 9700 PCR analyzer and 7500 Fast PCR analyzer calibration.Compared with the thermocouple measurement method,the temperature accuracy and temperature uniformity are greatly improved after using the temperature calibration of PCR analyzer device.
