Polymerase Chain Reaction, which is called PCR in abbreviation,is a kind of molecular level of biology technology, producing million s ofcopies of specific sequences of DNA in vitro. The PCR instrument is aninstrument which can imitate the temperature environment of the body for thePCR. At present, PCR technology has been widely applied in life scienceresearch, clinical diagnosis,biochemical analysis, drug analysis and in fieldwhere need gene cloning and manufacturing mutations.This paper discusses the temperature test and calibration of the PC Rinstrument. A temperature calibration system based on multi-sensortechnology is introduced, and the expanded uncertainty of the s ystem intemperature calibration around0.03℃. LabVIEW is chosen as thedevelopment software of the system and the calibrated results are reportedautomatically. Then study the temperature calibration specification of thePCR instrument on the basis of the calibration experiments. The main work isas follows:(1) The paper summarizes the principle of PCR,the thermal cycling ofPCR instrument, the influence of temperature on PCR and the researchsituation about PCR temperature detection home and abroad. Then theresearch contents and methods are put forward.(2). The paper studies about the trend of temperature distribution in theblock of PCR instrument through the finite simulated results of the steadytemperature field of the PCR instrument and suggests a multipoint-real liquidscheme for temperature calibration of PCR instruments. Based on weightedmean and group estimation,a multi-sensors data fusion method is applied indata processing. The result seems to be fine.(3). The hardware platform of the temperature calibration system of thePCR instrument is designed. Automatic calibration software based onLabVIEW is developed to achieve the calibration of the various parameters such as accuracy、uniformity and heating or cooling rate of the temperature,generate detection report and so on. Besides, Calibrate experiments of thePCR instruments are carried out in the laboratory, and then the calibrationresults are discussed. After that,the uncertainty of the measuring system andthe value error are analyzed respectively.(4). Finally, the paper introduces a simple application of the calibrationsystem with discussing the best annealing temperature of the cotton. PCRinstrument calibration specification is studied.
