Objective Agarwood is a kind of resin-containing wood of Aquilaria sinensis(Lour.)Gilg,and it is also a traditional and precious chinese medicinal material with a long medicalhistory.The existing literature reports are mainly used isolation and identification of endophytic fungi from agarwood to promote the agarwood formation from Aquilaria sinensis trees,but there is no report that the total enzyme produced by the endophytic fungi can directly catalyze the Aquilaria sinensis.In this paper,isolation and identification of endophytic fungi from agarwood study their ability to produce enzymes that catalyze Aquilaria sinensis(Lour.)Gilg with the change of the final chemical composition by modern separation and analysis strategies and biotechnology.Methods The endophytic fungi of agarwood were isolated and purified from Chen Xiang block,fermented and cultured to prepare total enzyme.The total enzyme was used to catalyze Aquilaria sinensis.HPLC was used to detect the change of the product after the total enzyme produced by endophytic fungus catalyzed Aquilaria sinensis.Results Twenty-one strains of agarwood endophytic fungi were isolated and purified,then fermented to prepare total enzymes.Each strain was cultured in a shaking table at 25℃ for 3~5 d with a fermentation volume of 24 L.The results showed that YZ-8 strain was the most enzyme-producing strain with 384.257 g;The least is BL(3)strain with 32.906 g.HPLC was used to analyze the ethanol extract of Aquilaria sinensis before and after the total enzyme catalyzed.The results showed that the chemical composition of Aquilaria sinensis changed significantly after catalyzed by total enzyme which produced from strains of YZ-8 and YZ-9.In addition,the chemical composition of Aquilaria sinensis changed to some extent after catalyzed by total enzyme which produced from strains of BL3(3)1(1),ML-1,YZ-1,YZ-3 and YZ-4.Seven endophytic fungi such as BL3(3)1(1),ML-1,YZ-1,YZ-3,YZ-4,YZ-8,YZ-9 were used for solid catalysis of Aquilaria sinensis.The ethanol extracts of strains before and after the catalysis of Aquilaria sinensis were analyzed by HPLC,the results showed that the chemical composition of Aquilaria sinensis was significantly changed after the strains of BL3(3)1(1)and YZ-1 were catalyzed.By using morphological and molecular biological methods,YZ-1 and BL3(3)1(1)were identified as Penicillium polonicum,YZ-8 as Arthrinium arundinis,and YZ-9 as Aspergillus sp.Conclusion The total enzymes produced by endophytic fungi of agarwood has catalytic effect on Aquilaria sinensis,which can change the chemical composition of Aquilaria sinensis significantly.This study laid a foundation for further research on the catalytic mechanism of the total enzymes produced by endophytic fungi of agarwood to catalyze on Aquilaria sinensis.
