Establishment And Application Of Chromatographic Fingerprints Of Agarwood | | Posted on:2019-04-13 | Degree:Master | Type:Thesis | | Country:China | Candidate:L L Shang | Full Text:PDF | | GTID:2371330548476668 | Subject:Applied Chemistry | | Abstract/Summary: | PDF Full Text Request | | Agarwood,as a rare spices and valuable ingredients of traditional chinese medicine,has high economic value due to a large number of aromatic and medicinal active ingredients.The agarwood chemical constituents are very complicated and concern many factors such as the time of agarwood,agarwood-induction technology,the growth of trees and the environmen.Traditional identification the material method more depends on experience which lacks scientific basis.Therefore,the development of commonly used scientific identification method based to its characteristic chemical constituent for protection and sustainable utilization agarwood,and the healthy development of agarwood industry is important.In this paper,wild agarwood and cultivated agarwood were selected as the research objects.And the HPLC(High Performance Liquid Chromatography)and GC-MS(Gas Chromatography-Mass Spectrometer)fingerprint chromatograms were established for the chromone and sesquiterpenes.On this basis,HPLC chromatograms of agarwood were established on specialty characteristic compound of 2-(2-phenylethyl)chromone.This chromatograms can be used to identify the authenticity of agarwood and distinguish the cultivated and wild agarwood.The main results are as follows :(1)The HPLC analysis method was established for chromone components.The HPLC chromatograms of 12 batches of agarwood showed that 5,6,7,8-tetrahydro-2-(2-phenylethyl)chromones(THPECs)was a common and high content component in wild and cultivated agarwood.And 5,6:7,8-diepoxy-2-(2-phenylethyl)chromones(DEPECs)and2-(2-phenylethyl)chromones(FTPECs)were the most diverse components wild and cultivated agarwood..Besides,the HPLC fingerprint of volatile components of agarwood could be used for quality analysis of agarwood.In addition,the HPLC chromatograms of chromone was was established.The characteristic peaks relative retention RSD were less than 2%,which showed good retention of reproducibility.It could be used for the quality analysis of the components of chromone.(2)The GC-MS analysis method was developed for volatile constituents of agarwood,including aromatic hydrocarbons,sesquiterpenes and chromone.The GC-MS chromatograms of 10 batches of agarwood showed that sesquiterpene and chromone were the main components of volatile compounds.Besides,the difference of sesquiterpenoids was more significant than chromone between wild agarwood and cultivated agarwood.Based on this,the GC-MS fingerprints of volatile components of agarwood could be established,and the characteristic peaks relative retention RSD(Relative Standard Deviation)were less than 2%.,which could be used for the quality analysis of the volatile components of agarwood.(3)Based on fingerprint analysis of chromone and volatile components of agarwood,the HPLC analysis method for the identification of agarwood could be established.Through 29 batches of HPLC chromatograms of wild and cultivated agarwood,HPLC Characteristic Chromatogram of agarwood was established.And the characteristic peaks relative retention RSD were less than 2%.The results of 15 batches of validation samples showed that the HPLC characteristic chromatogram of agarwood was accurate and effective for the identification of agarwood.Therefore,the HPLC characteristic chromatogram of agarwood showed good applicability for agarwood,which could be used for the quality control of agarwood.Based on the HPLC characteristic chromatogram of 48 batches of of agarwood,Fisher discriminant model of wild agarwood and cultivated incense was established.Based on Fisher discriminant analysis,the classification accuracies of wild agarwood and cultivated agarwood within three years all reached 100 %.Therefore,this model can be used for the identification of wild agarwood and cultivated agarwood within three years. | | Keywords/Search Tags: | Agarwood, HPLC, GC-MS, Fingerprint, Identification | PDF Full Text Request | Related items |
