Pesticide poisonings are frequent to people’s daily life and living environment pollution bring greater risks,so the high selectivity,natural degradation and the environment does not produce serious pollution of the green pesticide is one of the current direction of pesticide research and development.A large number of side effects of chemical pesticides such as pesticide residues and drug resistance,can be far better than the inhibition of crop diseases and pests,the supply of terrestrial plant antimicrobials from natural products is relatively inadequate,and the adaptability of terrestrial plants and the use of terrestrial plant resources have been used in excess,continue to find more active new pesticides have a certain degree of difficulty,it is far less than the special habitat of marine resources on the plant pathogen inhibitory activity was significant,marine fungal secondary metabolites have great potential in combating crop diseases,this is the starting point of this research: do isolation and purification and activity screening of marine endophytic fungi with Qingdao sea area rich marine resources,study on the bio-active components of the special sStrain for secondary metabolites.Fermentation was carried out using 4 different nutrient medium,four marine endophytic fungi were screened by comparin of productivity,metabolic abundance and biological activity,it turns out : in terms of productivity(secondary metabolites),RICE medium is more productive,MH1 medium follows,other secondary metabolic volume is small.In the metabolic product richness(TLC thin plate chromatography)screening,we found that differences in medium composition and culture time were found,which has a greater impact on the secondary metabolites of fungi,Secondary metabolites of CCH-3 and 03-A RICE medium,10-A MH1 medium are more abundant.Biological activity screening mainly includes two aspects: bacterial inhibitory activity and fungal inhibitory activity: GPYM secondary metabolite active medium CCH-3 produced the strongest by screening of four bacterial antibacterial activities,the maximum inhibition zone of BS is 28mm(positive control is 24mm),the maximum inhibition zone of SA is 25mm(positive control is 23mm),RICE medium follows,other three strains moderate antibacterial activity(inhibition zone is about 10?18mm);do the antimicrobial activity of four agricultural pathogenic,we found that agricultural fungi are barely amplified due to CCH-3 strain’s rapid growth and lush,which has a significant inhibitory effect.Endophytic fungi(CCH-3)from Chondria crassicaulis can be seen as the object of further study due to growth morphology singular,secondary metabolic rich,significant activity.The marine endophytic fungus CCH-3 was identified by using molecular biology means,it uses RICE culture medium scale fermentation,we get the extract and perform a VLC gradient elution,the antimicrobial activity of different polar components was tracked,the Fr.5-Fr.12 component has better activity,the inhibition zone of Bacillus subtilis(BS)with Fr.9 was 21 mm;staphylococcus aureus(SA)inhibition effect is better which inhibition zone is 28mm;in order to investigate the interference and influence of homologous fungi CCH-1 on its metabolites,CCH-3 and CCH-1 were mixed and fermented at the same time,the components with higher antibacterial activity were separated and purified by various column chromatography such as silica gel,gel,reverse phase,preparation plate and so on,more than 20 monomer compounds were obtained,four compounds have been identified by structural identification by modern spectroscopic techniques.In this paper,the isolation and purification of marine endophytic fungi,optimization of training to start with activity Isolation and Structural Identification of Secondary Metabolites,the two endophytic fungi of CCH-3 and CCH-1 were systematically studied,which provides a scientific basis for the development and utilization of marine microbial agricultural resources and provides the necessary and adequate basis for the protection for the new pesticide research and development.Monomeric compound AS-7 has a good suppression effect for BS(inhibition 45 mm,positive control 32mm),SA(inhibition 47 mm,positive control 33mm)、BT(inhibition 40 mm,positive control 35mm)、EC(inhibition 40 mm,positive control 33mm)、PA(inhibition 38 mm,positive control 35mm)、XC(inhibition 42 mm,positive control 32mm),inhibition zone than chloramphenicol(positive control)higher,this shows that the monomeric compound AS-7 has a strong antibacterial activity,worthy of further study.
