Monitering by content of orientin and vitexin and anti-oxidation activity,Trollius ledebouri Reichb antioxidant active components were extracted,purified,separated and identified.The purified substance were added to a cosmetics to expiore its anti-aging effect by human body function evaluation.The contents were as follows:(1)The Separation and purification were used to isolate by macro-resin method,polyamide chromatography,hydroxypropyldextran gel chromatography,etc.,And structure estimation and identification by Mass Spectrometry and Nuclear Magnetic Resonance.The results showed that the obtained monomer compounds were orientin,vitexin,quercetin and luteolin.Taking the amount of the obtained compound and the scavenging of DPPH free radicals as indicators,the best index components of orientin and vitexin were finally determined.(2)Through the single factor experiment and orthogonal experiment,the extraction process and purification process of orientin and vitexin in Trollius ledebouri were studied.The results showed that the optimal extraction process was: 60% ethanol,the ratio of material to liquid was 1:33,and extraction was 140 min.The best purification process is AB-8macroporous resin with a loading concentration of 10 mg/m L,a loading p H of 4,a loading flow rate of 10 m L/min,and a desorption solvent of 50% ethanol.(3)Its Antioxidant Activity was assessed by DPPH free radical experiment,ABTS free radical experiment and iron ion reduction experiment.The results indicate that: when the sample concentration is 0.038 mg/m L,the DPPH free radical scavenging rate is higher than50%,and the effect is equivalent to VC,which is 2.4 times higher than that before purification;In ABTS free radical scavenging experiment Among them,the samples of each concentration have a better scavenging effect on ABTS radicals,which is 4 times higher than that before purification.In the experiment of iron ion reduction,at 1 mg/m L,the reducing ability was nearly 3 times higher and showed a good iron ion reducing power.(4)Through the research on the cosmetic formula and preparation process,the extract is added to the cream skin care product to explore the effect on anti-oxidation.The results showed that after two months of using the cream,the skin moisture,wrinkles and thickness of the human body were significantly improved,and the anti-aging effect was exhibited to some extent.
