| The body’s free radical level is imbalanced under oxidative stress,and reactive oxygen species generated can attack the biological macromolecules,causing cellular damage,tissue damage,etc.,inducing a series of diseases and posing a great threat to health.Therefore,in order to prevent oxidative damage to the body,the anti-oxidation interventions from vitro is particularly important.Plant polysaccharides have attracted extensive attention as an active biomacromolecules.The Physalis alkekengi L.has a long history of planting in China,containing various bioactive components.Therefore,the Physalis alkekengi L.are selected as raw materials for polysaccharide extraction in order to extract bioactive polysaccharide to intervene oxidative stress,which has great significance for improving the antioxidant capacity of the body.With the Physalis alkekengi L.fruits selected as raw materials,and the extraction rate of Physalis alkekengi L.polysaccharide as the indicator,the extraction process was optimized by orthogonal test.The acid-added enzyme hot water extraction method was used to extract the Physalis alkekengi L.polysaccharide,in which the extraction temperature is 45℃,the extraction time is 2 h,the ratio of material to liquid is 1:35,the amount of citric acid is 3%,and the amount of cellulase is 3%.The evaluation on antioxidant activity in vitro of crude polysaccharides indicates that crude Physalis alkekengi L.polysaccharides have scavenging activities of DPPH radical,hydroxyl radicals and ABTS radicals.After further purification of phytoplasmic polysaccharides,the polysaccharide component Phy-1 was obtained by using cellulose anion exchange column,the polysaccharide content of which is 96%.FT-IR analysis revealed that it has a characteristic absorption peak of the polysaccharide and contains a pyranose ring structure.The molecular weight of Phy-1 was determined by gel permeation chromatography and found that Mass average molar mass was mainly distributed around 110 and 22 kDa.The research on the in vitro antioxidant activity of the purified polysaccharide indicates that Phy-1 is significantly higher than that of the Physalis alkekengi L.crude polysaccharide.After Phy-1 was digested in vitro,the small molecular weight of polysaccharides increased.The Mass average molar mass of around 110 kDa and 22 kDa polysaccharides was shifted to 108 kDa and 16 kDa,indicating that the part of larger molar mass polysaccharides has higher antioxidant activity.In the vivo antioxidant experiment,the antioxidant enzyme activity of mice feeding with Physalis alkekengi L.polysaccharides in the experimental group was higher than that of the model group and the malondialdehyde of the mice in diabetes model group was higher than that of the control group,indicating that Physalis alkekengi L.polysaccharides have antioxidant activity in vivo,could increase the activity of antioxidant enzymes and assist in scavenging free radicals in vivo. |
