Objective:The optimal preparation process of Xinxiang Nanoemulsion Spray was optimized by means of multi-index comprehensive evaluation combined with pharmacodynamic indexes to study its pharmacodynamic effect on allergic rhinitis(AR)mice and explore the mechanism of drug action.Methods:(1)The optimum extraction process of volatile oil was determined by L9(3)4 orthogonal experiment with the extraction amount of volatile oil as the index.With the inclusion rate and recovery rate as indexes,the optimum inclusion process of volatile oil was determined by L9(3)4 orthogonal experilent.Gas chromatography-mass spectrometry(GC-MS)was used to analyze the chemical constituents of volatile oil before and after inclusion.Taking the multi-index comprehensive score as the index,the optimal water extraction and alcohol precipitation technology of traditional Chinese medicine was determined by L9(3)4 orthogonal experimental design.(2)48 BALB/c mice were randomLy divided into 6 groups:Blank group,Model group,Positive drug group,Group A,Group B,and Group C,with 8 mice each group.The AR model of mice was established by OVA sensitization.After 1 week of treatment,scores of nasal behavioral symptoms,body mass,thymus and spleen mass and index,pathological changes of nasal mucosa,serum total Immunoglobulin E(IgE),ovalbumin-specific IgE antibodies(OVA-sIgE),and thymus factors interleukin-4(IL-4),interleukin-17(IL-17)and interleukin-3 3(IL-3 3)were observed.Results:(1)The optimal extraction process of volatile oil is:add 6 times the amount of water to the medicinal material,soak for 1h,decoction for 4h each time,and extract twice.The two volatile oils are combined into the total volatile oil.The optimal inclusion process of volatile oil was:saturated water solution method,the ratio of volatile oil to cyclodextrin was 1:6,the inclusion temperature was 40,and the inclusion time was 3h.After gc-ms detection and analysis,there was no significant difference in the main components of volatile oil before and after inclusion.The best extraction process of volatile oil is:add 6 times the amount of water to the medicinal material,soak for 1h,decoction for 4h each time,and extract twice.The best inclusion process of volatile oil is:using a saturated solution method,the ratio of volatile oil to?-cyclodextrin(?-CD)is 1:6,the inclusion temperature is 40?,and the stiring time is 3h.GC-MS analysis showed no significant difference in the main components of volatile oil before and after inclusion.The optimal extraction process of water extraction and alcohol precipitation is as follows:add 10 times of water,decoction for 3h,extract twice,add 95%ethanol to make the alcohol content reach 65%,and let the alcohol sink overnight(2)Pharmacodynamic experiments showed that compared with the model group,the symptoms of mice in each treatment group were significantly improved(P<0.05),and the thymus index and spleen index were significantly decreased(P<0.01).There was no obvious glandular hyperplasia and eosinophil infiltration in the nasal mucosa of the treatment groups.Compared with the model group,the serum OVA-sIgE in each treatment group was significantly reduced(P<0.05),the total IgE was also reduced,and the thymus cytokines IL-4,IL-17,and IL-33 were all reduced to varying degrees.Conclusion:After optimization,the extraction process of volatile oil from Xinxiang Nanoemulsion Spray produced a high oil content and the volatile oil inclusion process was stable and feasible.The technology of water-extraction and alcohol-precipitation has reached the requirements of the highest transfer rate of index components and stable quality.The Xinxiang Nanoemulsion Spray has a definite effect on AR mice,which may exert the anti-AR effect by inhibiting the level of thymic cytokines.The optimized volatile oil extraction process of the pungent nasal spray nanoemulsion has a higher oil yield,and the volatile oil inclusion process is stable and feasible.The water extraction and alcohol precipitation process has reached the requirements of the highest index component transfer rate and stable quality.Xinxiang nasal spray nanoemulsion is effective in AR mice,which may exert anti-AR effects by inhibiting thymic cytokine levels.
