Synthesis And Characterization Of Gadolinium Glycyrrhetinate Complex And Its Application In The Treatment Of Rhinitis Rats

Allergic rhinitis(AR)is a chronic inflammatory disease of the nasal mucosa,and with the development of society and the acceleration of industrialization,the number of AR patients is increasing year by year.The current clinical use of AR is effective but accompanied by side effects,therefore,the development of more effective AR drugs has become a focus of attention.Glycyrrhetinic acid(GA)has anti-inflammatory and anti-viral activities,while lanthanide metals have anti-inflammatory and anti-bacterial and anti-tumor effects.We combined gadolinium,a lanthanide metal,with GA to form the complex Gd-GA to investigate the therapeutic effects of Gd-GA on AR.The work of the paper is summarized as follows:1.The concept of AR and the important inflammatory factors involved in the pathogenesis are introduced,the pharmacological activities and effects of GA as well as lanthanide metals are outlined,and the significance as well as prospective of Gd-GA for the treatment of AR are reviewed.2.Synthesis as well as structural characterization of Gd-GA.By using elemental analysis,single crystal diffraction,mass spectrometry,infrared spectroscopy,thermogravimetric analysis and quantitative calculation-assisted simulation,the structure of Gd-GA was finally determined,with Gd bound to GA in 1:3 and with 18 lattice waters.3.The rat AR model was established,and the rat AR model was successfully established by PCA experiment,and then the methods of behavioral research,HE staining,and mast cell staining were used to explore the difference between eosinophils and mast cells in the blank group,AR group,and positive group.The difference between the control group and the Gd-GA group and the effect of Gd-GA on the behavior of AR rats and the histopathological morphology of the nasal and intestinal mucosa.Aggregating with mast cells and proliferating glands in the intestinal mucosa,Gd-GA plays a therapeutic role in the physiological regulation of AR pathogenesis.4.The rat AR model was established,and the expression of cytokines IL-4,IL-5,IL-10,and IFN-y in tissues was used as the curative effect index of Gd-GA on rat AR by immunohistochemical method.The immune mechanism of Gd-GA on rat rhinitis model was explored.The results showed that Gd-GA significantly decreased the expressions of IL-4,IL-5 and IL-10 in AR rats,while the content of IFN-y was significantly increased,Gd-GA may relieve symptoms such as nasal congestion,runny nose,and sneezing in AR rats by regulating mast cells and their downstream inflammatory pathways.
