Cinnamomum camphora seed kernel oil(CCSKO)is rich in medium-chain oils,which has the functions of reducing body fat deposition and rapid energy supply.It has high potential application value,but the content of its unsaturated fatty acids is extremely low.Incorporating EPA and DHA into CCSKO can increase the nutritional value of CCSKO and provide essential fatty acids.In this study,CCSKO and ethyl ester fish oil were used as substrates to prepare structured lipids containing EPA/DHA through the transesterification reaction catalyzed by immobilized lipase.There lipases were used,it was determined that the immobilized lipase of Novozyme 435 with higher EPA/DHA structured lipids yield,was the best catalyst.Single factor experiment and orthogonal experiment were used to study the influence of four factors on the transesterification reaction,such as the substrate molar ratio(CCSKO/ethyl ester fish oil),reaction temperature,reaction time and enzyme loading.The results showed that the optimal conditions were:substrate molar ratio(CCSKO/ethyl ester fish oil)1:1,reaction temperature 60?,reaction time 8 h,and enzyme loading 10%.The maximum EPA/DHA incorporation yield was 24.57%in this optimized condition.Molecular distillation was used to separate and purify the transesterified crude product obtained after the scale-up experiment.Gas chromatography analysis revealed that the total saturated fatty acid content in the product was 75.85%,mainly including capric acid(43.34%)and lauric acid(30.67%).The total unsaturated fatty acids were 24.15%,the contents of long-chain unsaturated fatty acids of EPA and DHA were 12.01%and 7.94%,respectively.Furthermore,the ratio of total saturated fatty acids and total unsaturated fatty acids was close to 3:1.The contents of saturated fatty acids unsaturated fatty acid in sn-2 position was 76.66%and 23.34%,respectively.It was mainly including EPA(11.72%)and DHA(7.13%).Triglyceride analysis showed that the main triglycerides in the product including CCC(8.98%),CC-EPA(11.07%),CCLa(32.89%),CC-DHA(6.71%),CLa-EPA(7.78%),CLa-DHA(5.99%)and MCC/ClaLa(13.91%),the purity of the product after purification reached 44.22%.By analyzed and compared the physicochemical properties and the antioxidant capacity in vitro of the oil,the results showed that the CCSKO and molecular distillation products met the relevant hygienic standards for edible vegetable oil in terms of acid value,saponification value,iodine value and peroxide value.CCSKO and molecular distillation products have a certain ability to scavenge DPPH,ATBS and OH free radicals,and the ability to chelate ferrous ions.Furthermore,the melting point of CCSKO and molecular distillation products was lower than the body temperature.In summary,CCSKO and this new structured lipids have great physicochemical properties and nutritional characteristics.There is great potential for development in the daily oil and fat and healthy food industries.
