This study is on the optimized selection of the parts processing process of Chinese traditional medicine Corydalis yanhusuo,mainly including the extraction process of dl-tetrahydropalmatine from Corydalis yanhusuo,and extraction process,drying process of Corydalis yanhusuo concrete,In this process which use of environmentally friendly solvents.Under the premise,a quality safety evaluation system for Corydalis yanhusuo extractum was established.In this study,the contents of six alkaloids,such as dl-tetrahydropalmatine,of the extraction of Corydalis yanhusu was determined by high performance liquid chromatography.The contents of 5 kinds of heavy metal elements were determined by atomic absorption spectrometry.the contents of two organochlorine pesticide residues in Corydalis yanhusuo extractum was determined by gas chromatography.Purpose of this study This research topic is to through“laboratory test”to explore optimum production process and conditions of Corydalis yanhusuo extractum,for standardization and production efficiency in the process of Corydalis yanhusuo extractum processing quality to provide the corresponding theoretical basis.Through the formulation of the quality safety evaluation system of Corydalis yanhusuo extractum,it provides reference for the quality consistency and product safety of the production of Corydalis yanhusuo extractum.The research contents and results are as follows.Part 1 Study on The Optimum Extraction Process of dl-tetrahydropalmatine by Supercritical CO2OBJECTIVE To study the optimum conditions of the extraction technology and condition of dl-tetrahydropalmatine.METHODS Contrast the extraction of dl-tetrahydropalmatine from Corydalis yanhusuo by Soxhlet extractor method,Ethanol Extraction Method,Microwave assisted Extraction,and Supercritical fluid extraction(SFE-CO2),in terms of the extraction of dl-tetrahydropalmatine and extract yield.The extraction technology of dl-tetrahydropalmatine from Corydalis yanhusuo was optimized by orthogonal experiment of Orthogonal experimental design L9(34).To extract the dl-tetrahydropalmatine in Corydalis yanhusuo by the SFE-CO2 and to measure the contents of dl-tetrahydropalmatine by HPLC.RESULTS It is concluded that the Supercritical carbon dioxide fluid extraction(SFE-CO2)method is the best method to extract the dl-tetrahydropalmatine.The optimal technology was as follows:choose the herbs’Particle size of 30 mesh number,the extraction pressure of 20 MPa at the extraction temperature of 45℃,with the modifying agent is the saturated Ca(OH)2 solution and the entrainer is 95%ethanol solution which the quality of 1.5times of the herbs.CONCLUSION The process is stable and feasible for the extraction of dl-tetrahydropalmatine from Corydalis yanhusuo.Part 2 Study on preparation process optimization of Corydalis yanhusuo extractumOBJECTIVE Based on the comparative study of the extract methods and concentrated drying method of Corydalis yanhusuo extractum.METHODS In terms of the extraction of dl-tetrahydropalmatine and total alkaloids and extractum yield.First,to study of two commonly used methods for the preparation of Corydalis yanhusuo extractum,Including percolation and heat reflux.Secondly,studied the preparation effect of acetic acid,ethanol and acetic acid of different concentration gradients and distilled water.finally,to screen the optimum process in 5 different drying methods,including Box-type drying、Freeze drying、Microwave drying、Infra-red drying and Vacuum belt drying.RESULTS The experimental results show that in the process of processing Corydalis yanhusuo extractum,use heat reflux method to prepare Corydalis yanhusuo extractum,and select a concentration of 20%acetic acid as extraction agent(solvent),using the vacuum belt drying method for enrichment and dry is the best way of processing Corydalis yanhusuo extractum.CONCLUSION This production efficiency of Corydalis yanhusuo extractum is the highest,and the process is stable and feasible.It can be used in industrial production.Part 3 Quality Safety of Corydalis yanhusuo extractumOBJECTIVE Based on the Corydalis yanhusuo of Chongqing kaizhou experimental base as a medicine source,use the best preparation technology of Corydalis yanhusuo extractum to carry out the production of"small test"in the laboratory.The quality safety monitoring and evaluation of the modified Corydalis yanhusuo extractum includes the content of the active ingredients in the extract of Corydalis yanhusuo,the residual quantity of heavy metals and the content of pesticide residues.Draft a draft quality safety evaluation system for Corydalis yanhusuo extractum.METHODS(1)HPLC(High performance liquid chromatography)was used to determine the content of six major alkaloids,such as tetrahydroxanthine in Corydalis yanhusuo extractum;(2)AAS(Atomic absorption spectrometry)was used to determine the content of 5kinds of metal elements,such as mercury,lead and chromium in Corydalis yanhusuo extractum;(3)the determination of DDT and 666 organochlorine pesticide residues in Corydalis yanhusuo extractum were determined by GC(gas chroma-tography).CONCLUSION Experimental results show that the content of alkaloid active ingredient,heavy metal and pesticide residues conform to the provisions of the quality and safety requirements of Corydalis yanhusuo and its’extractum in National standards for food safety of the People’s Republic of China,Green standards of medicinal plants and preparations for foreign trade and economy and The Zhejiang local standard for the production technology of yanhusuo.
