ObjectiveThe seeds of Abelmoschus manihot(L.)Medic.were studied from the aspects of traits,identification,inspection items,extracts and content determination.At the same time,structural identification,compositional analysis and systematic methodological studies were carried out.Thus,the method for determining the content of fatty acids in the seeds of Abelmoschus manihot(L.)Medic.was established,and the draft quality standards and drafting instructions were prepared to provide a basis for the establishment of quality standards.MethodsThe traits of the seeds of Abelmoschus manihot(L.)Medic.were summarized and summarized by eye observation,hand touch and nasal smell.Microscopic identification was carried out by microscopy and SEM,and TLC identification was carried out,and IR was performed by FTIR.According to the 2015 edition of the Chinese Pharmacopoeia,the seeds of Abelmoschus manihot(L.)Medic.were used to determine the moisture,total ash,acid-insoluble ash,extracts and heavy metals.At the same time,the structure was identified by GC-MS.And carry out component analysis,and at the same time,systematic methodological study on its fatty acid components by GC method.The determination of the content of palmitic acid,oleic acid and linoleic acid in the seeds of Abelmoschus manihot(L.)Medic.ResultsThe seeds of Abelmoschus manihot(L.)Medic.were observed and measured,and the traits were described objectively and practically in combination with the support of relevant literature.Microscopic identification and TLC showed that the microscopic characteristics were obvious,the sample had good resolution and the spots were clear;A large amount of long-chain fatty acids are present in the medicinal materials of IR.The moisture content of the 10 batches of the seeds of Abelmoschus manihot(L.)Medic.is in the range of 9.99% to 12.14%,with an average of 10.78%;the total ash content is in the range of 4.75% to 5.67%,with an average of 5.18%.The acid-insoluble ash content is in the range of 0.18%~0.37%,the average value is 0.25%;the water-soluble extract content in the range of 18.63%~23.58% is determined by hot dip method,and the average content is 20.75%;The content of heavy metals in the medicinal materials is low,so it is not included in the standard.Six fatty acids were isolated by GC-MS,which were methyl palmitate,methyl stearate,methyl 8-oleate,methyl oleate,methyl linoleate and cis 9,10-methylundecane.Methyl ester.The normalized concentration of methyl palmitate,methyl oleate and methyl linoleate is 94.14%,so the three components can be selected as the content determination index.The results of the methodology showed that methyl palmitate,methyl oleate and methyl linoleate were 0.527~2.633mg/mL(r=0.9999),1.099~3.297mg/mL(r=0.9998)and 1.515~3.535 respectively.There was a good linear relationship in the range of mg/mL(r=0.9999);the average recoveries were in the range of 98.31%~101.13%,97.91%~101.48% and 98.42%~101.01%,respectively,and the RSD range was 0.95%~2.78%.The contents of palmitic acid,oleic acid and linoleic acid in 10 batches of Astragalus membranaceus were between 5.53%~6.28%,7.74%~16.19% and 10.52%~12.57%,respectively.The total fatty acids ranged from 24.49% to 33.25%.The value was 29.26% and the RSD was 10.10%.ConclusionsIn this paper,the seeds of Abelmoschus manihot(L.)Medic.were studied from the aspects of traits,identification,examination items,extracts and content determination.The structure identification of Astragalus membranaceus was carried out by GC-MS,and the composition analysis was carried out to provide a basis for the selection of the index components under the content determination.A systematic methodological study of the seeds of Abelmoschus manihot(L.)Medic.was carried out by gas chromatography.Establish a multi-index content determination method of palmitic acid,oleic acid and linoleic acid.The method is specific and reproducible,and can be used for quality control of the seeds of Abelmoschus manihot(L.)Medic.
