| Natural product herbal medicines have a complex chemical composition,in which the active ingredients in the herbs may vary due to factors such as growth age,growth environment,and harvesting time.In recent years,a major area of research on herbal medicines is the analysis of the active chemical components in herbal medicines,however,the uneven quality of herbal medicines can seriously affect their clinical application.If the quality standards of herbs are studied and their limits are defined,herbs can be measured and controlled more precisely and scientifically.Artemisia sieversiana,which belongs to the genus Artemisia in the family Asteraceae.Artemisia sieversiana is widely distributed in China and has abundant plant resources.The volatile oil of Artemisia sieversiana has remarkable biological activity and can be used as raw materials for the manufacture of natural antioxidants,fungicides,spices and other products with the advantages of high efficiency and low toxicity.At present,the research on Artemisia sieversiana medicine is less in-depth and the quality standard of herbal medicine is not perfect,which makes it difficult to evaluate the quality of Artemisia sieversiana medicine scientifically.In this study,the volatile oil of Artemisia sieversiana was found to have good biological activity during the preliminary literature screening,so the plant was selected as the raw material for the process optimization and antioxidant activity study of the volatile oil of Artemisia sieversiana.In order to systematically and comprehensively control the quality of Artemisia sieversiana,the quality standard of Artemisia sieversiana was initially established.The main research contents are as follows:1.It was demonstrated that the extraction of volatile oil from Artemisia sieversiana can be more effective by using salinization-assisted hydrodistillation method.The effect of each factor on the extraction rate of volatile oil of Artemisia sieversiana was investigated by single-factor experiments.9 experiments with 4 factors and 3 levels were designed by orthogonal experimental method to optimize the extraction method of volatile oil of Artemisia sieversiana,and the best extraction process was obtained as follows: materialliquid ratio of 1:8,heating temperature of 160℃ in electric heating jacket,extraction time of 8h,and concentration of Na Cl solution of 7%.The volatile oil content of different batches of Artemisia sieversiana was measured in the range of 0.55%~1.10% using volatile oil as the index component2.The in vitro antioxidant activity of the volatile oil of Artemisia sieversiana was investigated by measuring the scavenging ability of DPPH radicals and ABTS radicals as well as the total reducing ability test using VC as positive control.The results showed that the volatile oil of Artemisia sieversiana had a high antioxidant capacity.3.Ten batches of Artemisia sieversiana from different regions of Xinjiang were collected,and the trait identification characteristics and powder microscopic characteristics of Artemisia sieversiana were observed and recorded,and the thin-layer chromatographic identification method of chlorogenic acid and the thin-layer chromatographic identification method of volatile oils in Artemisia sieversiana was established.At the same time,the moisture,dilute ethanol leachate and total ash of Artemisia sieversiana were studied,and the limits were set according to the results,with the provisional moisture not higher than 11%;total ash not higher than 10%;acid insoluble ash shall not exceed 1.88%;dilute ethanol leachate not less than 10%.The content of chlorogenic acid in different batches of Artemisia sieversiana was measured by HPLC using chlorogenic acid as an index component.All RSDs of precision,reproducibility and stability tests were lower than 3.00%.Measured ten batches of Artemisia sieversiana seeds chlorogenic acid content of 0.4476mg/g~1.3200mg/g.Tentative determination of the herb chlorogenic acid content of not less than 0.35mg/g.4.Ten batches of Artemisia sieversiana samples were examined by HPLC.Ten sets of common peaks were selected,and chlorogenic acid was used as the standard peak,and the relative peak areas and relative retention times of the common peaks were used to calculate the RSDs for the methodological investigation.The results of the systematic cluster analysis of the ten batches of Artemisia sieversiana were classified into three major categories using SPSS 27 software.The fingerprints established in this way can provide a preliminary reference basis for the quality control of Artemisia sieversiana. |
