Study On Weight-loss Function Evaluation And Mechanism Of Seven Materials Including Banana Powder

According to the research results,green bananas contain mainly natural resistant starch,which has the function of controlling body weight and regulating intestinal flora.The purpose of this paper was to evaluate and compare the weight-loss function and mechanism of three kinds of banana powder and four kinds of weight-loss materials on the market through animal experiment and they were dajiao pulp powder(Musa ABB),dajiao powder with skin(Musa ABB),cavendish pulp powder(Musa AAA),corn starch,resistant dextrin,konjac powder and L-carnitine,respectively.The concrete research contents and results are as follows:(1)The nutritional obese rats as the research subjects were used to establish the nutritional obesity model and orlistat was selected as the positive medicine.Seven kinds of test substances were intragastrically administered at a dose of 2.5 g/(kg BW·d),and no significant changes were found in daily food intake and wet weight of organs.Dajiao pulp powder and konjac powder had better effect on reducing body weight of obese rats,at the end of the sixth week,compared with the obesity control group,the body weight in the above experiment groups decreased by 7.20% and 2.86% respectively.Dajiao pulp powder and konjac powder had better effects on reducing body fat content in obese rats and when compared with the obesity control group,the body fat in the dajiao pulp powder and konjac powder group was decreased by 37.15% and 41.73% respectively.High fat diet can increase liver weight in the rats,and liver wet weight was reduced significantly after the intervention of dajiao pulp powder and konjac powder.According to the observation results of rat liver slices,it was found that the order of weight loss function of seven materials was as follows(from better to worse): Dajiao pulp powder,konjac powder > dajiao powder with skin,resistant dextrin,corn starch > cavendish pulp powder and L-carnitine.(2)Blood biochemical and hormone indexes were measured by biochemical automatic instrument and ELISA kit.It was found that dajiao pulp powder,konjac powder and cavendish pulp powder had better effects on reducing blood sugar and lipid.Dajiao pulp powder,konjac powder,corn starch,resistant dextrin and cavendish pulp powder had better liver protection effect and can significantly improve the liver dysfunction in obese rats.Obesity induced by high-energy diet was related to the increase of serum insulin and leptin,and the decrease of serum ghrelin and adiponectin was appeared in obesity rats.Banana powder could control the weight of obese rats by decreasing the levels of insulin and leptin in the serum of obese rats,increasing the levels of ghrelin and adiponectin,alleviating the symptoms of hyperglycemia and hyperlipidemia caused by obesity,among which dajiao pulppowder was better than dajiao powder with skin and cavendish pulp powder.(3)The composition and structure of intestinal flora in rats were determined by 16 S rDNA gene sequencing method,and the content of short-chain fatty acids in rat feces was quantitatively determined by method of high performance liquid chromatography(HPLC).The results of macrogenomics showed that compared with the obesity control group,intestinal flora abundance of obese rats after the intervention of resistant dextrin and dajiao pulp powder was higher.At the level of phylum,Firmicutes and Bacteroidetes were the dominant phylum.At the level of genus,compared with the obesity control group and normal control group,corn starch could proliferate the bacteria of Verrucomicrobiaceae significantly,dajiao pulp powder could proliferate the bacteria of Coprococcus-2 significantly,and the content of Lachnospiraceae bacteria in the resistant dextrin group was remarkable.As we all known the above three species belong to beneficial bacteria which could produce more metabolites such as acetic acid,propionic acid and butyric acid.The results of HPLC showed that the content of SCFA in rat feces increased more after the intervention of resistant dextrin,dajiao pulp powder and corn starch,followed by cavendish pulp powder,dajiao powder with skin and konjac powder,while L-carnitine had no significant effect.Therefore,the effects of different interventions on regulating intestinal flora structure and improving metabolites were as follows: resistant dextrin,dajiao pulp powder,corn starch > cavendish pulp powder,dajiao powder with skin,konjac powder > L-carnitine.
