Objective:To develop the Sandouyin Beverage on the basis of the Sandouyin(black beans,mung beans,red beans)created by the ancient famous do ctor Bian Que,which has the effect of clearing away heat and detoxifying,activating blood circulation,nourishing the liver and moistening the lungs,moistening dryness and promoting fluid.The beverage was expected to be good in taste,easy in take and accepted by public easily.Methods:1 To develop the Sandouyin Beverage:(1)Method of determination of effective substance:the content of total polysaccharide was analyzed by the sulfuric acid-phenol method,and the content of total polyphenols was analyzed by the Folin-Ciocalteu chromatometry.(2)To optimize the water extraction process of Sandouyin:Using extraction times,water/solid ratio,extraction duration as response factors,the extraction content of total polysaccharide and total polyphenol as response value,based on single factor test,reponse surface method was used to obtain the optimal extraction process of Sandouyin.(3)To compare the antioxidant activity of the products:Antioxidant activities of the Sandouyin original extract and suitable extract was estimated by DPPH·and hydroxyl radicals.(4)To determine the blending process of Sandouyin:Determine the ingredients of Sandouyin Beverage by sensory evaluation,and determine the sterilization conditions and color protection conditions based on microbiological index,browning index and efficacy index.(5)To determine the shelf life of Sandouyin:Under the storage conditions of 50℃,24℃,37℃,the variety in Sandouyin Color protected group and unprotected group of microbial indicators,physical and chemical indicators and efficacy indicators were determin ed during the 90-day storage period.2 To develop the extraction process of Dietary Fiber from Soybean Dregs:Using acid extraction water/solid ratio,acid extraction time,acid extraction temperature,acid extraction pH as response factors,the extraction content of dietary fiber from Sandouyin soybean dregs a response value,reponse surface method was used to obtain the optimal extracti on process of dietary fiber from Soybean Dregs based on single factor of acid extraction test.Under the optimum conditions of acid extraction,Using alkali extraction water/solid ratio,alkali extraction time,alkali extraction temperature,alkali extraction pH as response factors,the extraction content of dietary fiber from sandouyin soybean dregs as response value,reponse surface method was used to obtain the optimal extraction process of dietary fiber from Soybean Dregs,based on single factor of alkali extraction test.3 Study on quality standard of Sandouyin Beverage:According to the seasoning process,sterilizati on process and color protection process of Sandouyin beverage,the formulation process standard was determined.According to the alter of physical and biological indicators,physical and chemical indicators and efficacy indicators during the shelf life,the shelf life and the change standards of various indicators of Sandouyin Beverage were determined.Results:1 To develop the Sandouyin Beverage:(1)There was a good linearity for Anhydrous Glucose in the range of 30.1 μg/μL-101 μg/mL(r=0.9992).The average recovery was 97.24%,RSD 1.74%(n=9).There was a good linearity for gallic acid in the range of 23μg/mL~69μg/mL(r=0.9992).The average recovery was 97.28%,RSD 1.47%(n=9).(2)The result showed that the optimal conditions were:the water/solid ratio of 1:18,the extraction time of 3,extraction duration of 2.5 hours.Under the optimal condition,the content of total polysaccharide was 242.9mg/g and total polyphenol was 8.86mg/g.(3)The raw water extraction had a DPPH scavenging rate of 72.9%and a hydroxyl radical scavenging rate of 95%.The suitable ext raction solution had a DPPH scavenging rate of 74.5%and a hydroxyl radical scavenging rate of 97%.The concentration of the extracting solution by eliminating DPPH·50%was 206.2μg/mL and hydroxyl radical 50%was 184μg/mL.When the content of total polysaccharide of 70.5μg/mL-1127.5 μg/mL and the content of total polyphenol solution of 2.6 μg/mL-40.5 μg/mL,the antioxid ant activity of the sample solution increased with the increasing of the content of tota 1 polysaccharide and total polyphenol.(4)The ingredients of Sandouyin Beverage was determined through the sensory evaluation:100 ml of Sandouyin extraction,2000 mg of white sugar and 75 mg of salt.The Suitable sterilization conditions were:sterilization temperature of 100℃,time of 10 min.The best color protection process was 0.3%D-isoascorbate.(5)It was found that physical and biological indicators,physical and chemical indicators and efficacy indicators of the color protection group and the unprotected color group were basically stable,under the storage condition of 5℃ during the 90-day storage process.Under the storage condition of 25℃,the indicators of the color protection group were basically stable;the colony was formed in the unprotecte d group for 80 days,and the efficacy index was decreased.Under the storage condition of 37℃,colony was formed after 60 days in the color protection group and the unprotected color group,and the physical and chemical indicator s and efficacy indexes were decreased.The decline rate of each indicator in the unprotected color group was greater than that in the color protection group.2 The Study of Extraction process of Dietary Fiber from Soybean Dregs:The optimal acid extraction conditions of dietary fiber from soybean dregs of Sandouyin were:the water/solid ratio of 1:25,acid extraction time of 1.5h,acid extraction temperature of 87 C,acid extraction pH of 4.65.The content of dietary fiber after acid extraction was 59%,which increased by 14.4%compared with that of raw soybean dregs.The optimal alkali extraction conditions of dietary fiber from soybean dregs of Sandouyin were:the water/solid ratio of 1:35,alkali extraction time of 4h,alkali extraction temperature of 56 C,alkali extraction pH of 11.8.The content of dietary fiber after alkali extraction was 75.7%,which increased by 30.1%compared with that of raw soybean dregs.3 The results of all examination items of Sandouyin Beverages were in compliance with relevant regulations.Conclusion:1 The development of the Sandouyin Beverage:(I)This determination method was simple,stable,accurate and reproducible.(2)The content of total polysaccharide and total polyphenol of Sandouyin were basically similar with the predicted values.The method was stable and feasible.(3)The raw water extraction and suitable extraction solution had good antioxidant activity.(4)Blending process of Sandouyin Beverage was stable and feasible(5)The shelf life of 25 C of Sandouyin Beverage was 3 months.The shelf life was longer.2 The Study of Extraction process of Dietary Fiber from Soybean Dregs:The content of dietary fiber of soybean dregs were basically similar with the predicted values,the process was stable and feasible.3 The proposed quality standard of Sandouyin Beverage was effective,stable and contro lab e.
