Wax gourd(Benincasa hispida(Thunb.)Cogn.)is one of the annual herbaceous plants of the genus gourd,mainly distributed in subtropical regions and cultivated in all regions of China.Wax gourd,as an important vegetable crop,which is abundant,inexpensive and nutritious,rich in protein,polysaccharides,vitamins and other substances.In recent years,it has been found that polysaccharide has anti-tumor,hypoglycemic,immune,antioxidant and anti-aging effects.At present,there are few studies on the nature relatively,structure and activity of wax gourd.In this experiment,the preparation and extraction process of wax gourd polysaccharides were optimized by using wax gourd as raw material,and the skin activity of the wax gourd polysaccharides and the improvement of skin in aging mice were studied We hope to get an efficient method for extracting the polysaccharides from wax gourd and explore the potential of the development and utilization of wax gourd polysaccharides in the field of daily cosmetics.The main results are as follows:1.The ultrasound-assisted extracted technique of wax gourd polysaccharides were studied in this paper.On the basis of single factor experiment,this work studied the effect of material liquid ratio,extraction time,and extraction power on the extraction yield of wax gourd polysaccharides.Then the optimal extraction conditions were obtained by the orthogonal experiment.The influence of ultrasonic extraction time and material liquid on the extraction rate of winter melon was significant(P<0.05).The ultrasonic assisted extraction technology conditions for the material liquid of wax gourd polysaccharide than 1:40,40 min ultrasonic extraction time,extraction power 380 W,the conditions of crude polysaccharide from wax gourd was at a rate of 13.15%,compared with the hot water extraction,yield increased by more than 50%,shorten the extraction time by 2 times.The ultrasonic assisted extraction could significantly improve the extraction efficiency of wax gourd polysaccharide.At the same time,the crude polysaccharide samples of the wax gourd were identified.The protein content of the polysaccharide was 12.58%and the sugar content was 54.83%.It was an acid polysaccharide containing the glucoside of the glucoside.2.The Skin care activity of wax gourd polysaccharides were studied in this paper.studies the activity of wax gourd polysaccharides.The wax gourd polysaccharides was characterized by studying its reactivity to DPPH,hydroxyl free radical,superoxide anion free radical,hygroscopicity,moisture retention and inhibition of tyrosinase activity.When the concentration of wax gourd polysaccharides was 5 mg/ml,the clearance rate of DPPH,hydroxyl radical and superoxide anion radical was 77.91%,99.64%and 74.48%respectively;In the range of 0-12 h,wax gourd polysaccharides has the ability of continuous water absorption,reaching the maximum value at 12 h,the moisture absorption rate is 10.5%.In the dry environment,when 12 h,the moisturizing rate of the wax gourd polysaccharides is 19.60%,the moisture retention is better than the pure water,lower than the glycerol,when glycerol is mixed with the wax gourd polysaccharides,there is a significant synergistic effect and the 12 h moisture content reaches 44.20%;when the polysaccharides concentration of the wax gourd reaches 2 mg/mL,The inhibition rate of tyrosinase was 64.39%.Therefore,we can see that the polysaccharides of wax gourd has good antioxidant activity and good moisture absorption,moisturizing,whitening and skin care effects.3.The effect of wax gourd polysaccharides on aging skin in mice were studied in this paper.The effects of wax gourd polysaccharides and establishment of aging model on skin moisture content,SOD activity,CAT activity and MDA content in aging mice were studied.Using the wax gourd polysaccharides solution for 6 weeks,the moisture content,SOD and CAT activity in the skin of aging mice were significantly improved,and the MDA content was reduced significantly.Wax gourd polysaccharides can remove cell free radicals,inhibit the skin cell oxidative damage,delay skin aging.
